Objective:To investigate the expression and significance of TNF-a in articular cartilage during limbs ischemia-reperfusion injury.Methods:Fourty SD rats were used and divideded into 8 groups at rand-om:sham-operatedgroup,ischemia group,ischemia-reperfusion groups(1,3,7, 14,30,60day).Each group has 5 rats. Rats limb ischemia-reperfusion model was established from Chengjianchang' method. The grouped rats were killed respectively. The articular cartilage were excised from the medial of the left keen joint. The pathological changes of articular cartilage were observed by HE stain and the expression of TNF-awas confirmed by immmunohist-Oche mistry technique.All dates were expressed as mean±SEM.In the end,we ana-lysed the outcomes of this experiments with SPSS 13.0.Vaue of P<0.05 were considered statistically signifycation.Results:1)Cartilage cells lightly swelled at 7days after reperfusion and got worse following the time pass.2)In sham operated group, TNF-a postive expression was weak and increased at ischemia 6h group.The positive expression of TNF-a peaked at 3d,7d and then began to decrease.But there were stilsignificant difference between IR 60d group and sham-operated group (P<0.05)Conclusions:1)Limb ischemia-reperfusion injury can cause the injury of articular cartilage. 2) TNF-a plays an important role in the injury of articular cartilage after limb ischemia-reperfusion.
