Objective:To observe whether the metformin can inhibit the proliferation of MCF-7 cells and investigate the synergistic effects paclitaxel liposome combine with metformin in MCF-7 cells.1.The MCF-7 cells were treated with metformin in different concentration respectively, then the growth inhibiting effects were observed at different time by MTT assay.2.The MCF-7 cells were intervened with metformin, which concentration was 0.03 mmol/L and 0.1 mmol/L, combined with the paclitaxel liposome in different concentration(30ug/mL,60ug/mL). The growth inhibiting effects are observed by MTT assay in order to assess the interaction between metformin and paclitaxel liposome.1.We found that the inhibition ratio of the proliferation was significantly increased by increasing the metformin concentration and prolonging the intervention time, and the effects were dose-dependent and time-dependent.When metformin concentration greater than 1.0mmol/L and intervened after 24 hours, concentration greater than 0.03mmol/L and intervene 48 hours, or concentration greater than 0.01 mmol/L and intervene 72 hours, the inhibition ratio was significantly higher compared with the control group (p<0.01),In the 24h group and the 48h group,we found there were significant differences of inhibition ratio, when the metformin concentration greater than 3.0 mmol/L (p<0.01); and in 72h group, the inhibition ratio was significantly increased at the metformin concentration greater than 1.0 mmol/L。At the same concentration, the inhibition ratio was significantly higher with the prolongation of intervened time. There were significant differences (p<0.05) between 24h and 72h group when the concentration of metformin great than 0.01 mmol/L.There were significant differences (p<0.05) between 24h and 48h group when the concentration of metformin great than 0.03mmol/L. There were significant differences (p<0.01) between 48h and 72h group when the concentration of metformin great than 1.Ommol/L.Correlation analysis showed the inhibition ratio of the metformin was increased in a dose-dependent and time-dependent manner.2. The proliferation inhibition on mcf-7 cells significantly increased intervened by the combination of metformin and paclitaxel liposome than either of the drugs alone.Those effects had interactive impact (F=4.005, P<0.05). When the MCF-7 cells were intervened by 60 ug/L paclitaxel liposome combined with 0. 1mmol/L metformin, the inhibition ratio was the highest.1.Metformin has dose-dependent and time-dependent anti-proliferative effects on MCF-7 cells in vitro.2.the effect of the proliferation inhibition induced by metformin combined with paclitaxel liposome present synergistic effects.
