Prokaryotic Expression Of Candida Albicans Heat Shock Protein 90 And Its Immunogenicity

Candida albicans and other Candida species are opportunistic yeast that cause both mucosal and systemic candidiasis. Fungal infection on the body damage is great, especially the systemic C.albicans infection.According to British scientists'statistics, in the use of antifungal drugs, the disease's mortality rate remained as high as 70%. Therefore, control of C. albicans is the best way to prevent infection, but vaccine development is to build the key to effective preventive measures, be able to develop an effective vaccine to combat Calbicans infection is very important. DNA vaccine with incomparable advantage is the third generation vaccine which follows pathogeny vaccine and subunit vaccnie.One C.albicans cell wall antigen, heat shock protein 90kDa (Hsp90), and its degradation 47 kDa,40 kDa fragment are recognized by serum antibodies. The C.albicans Hsp90 is an important target for protective antibodies in disseminated candidiasis of experimental mice and humans. Matthews and others used the patients'serum and identified the epitope map of C.albicans Hsp90.They found three common epitopes which can react with the patients' serum,thereby they drew out the continuous epitopes of Hsp90 C end. Namely, epitope C,epitope B and epitope H.According to experiment,they proved that only epitope C and epitope B have immunogenicity.In this study, we selected the Hsp90 gene sequence that can encod the epitope C,epitope B and epitope H,and then constructed a prokaryotic recombinant plasmid named pET28a (+)/ Hsp90.And we exsspressed the gene in prokaryotic cells,according to Western tests we proved the immunogenicity of the recombinant protein.Mice immunized with the purified protein can generate a corresponding immune response,and then produce antibodies.This experiment further validated the immunogenicity of the recombinant protein.This laid the foundation of the research in C.albicans Hsp90 DNA vaccine.
