Objective:Through detecting of the bone marrow megakaryocyte membrane standardized count, T lymphocyte subsets in patients with idiopathic thrombocytopenic purpura, to explore the bone marrow megakaryocytes standardized count, T lymphocyte subsets and the clinical significance of operation for clinical standardization and diagnosis.Method:(1) Using traditional and standardized counting method detected the megakaryote number of 198 patients with ITP and 20 controls.(2)The percentage of T lymphocyte Subsets in 64 ITP patients and 31 healthy controls were tested by flow cytometry.Result:(1) Compared with the controls, the traditional and standardized counting of megakaryocyte in ITP patients were significantly increased (P<0.05). There was no apparent difference between the traditional and standardized counting of megakaryocyte with the patients or controls in ITP(P>0.05). After standardized counting of megakaryocyte, the CV value decreased compared to the traditional counting of megakaryocyte.(2) T lymphocyte subsets, the percentage of CD3+ T lymphocyte and CD4+ T lymphocyte and the ratio of CD4+/CD8+ in patients with ITP was 60.88±14.59%, 28.41±10.55% and 1.18±0.59% respectively, that were notably decreased than those healthy controls which of control group were 69.89±6.43%,35.38±5.05% and 1.64±0.29% respectively (P<0.05). The percentage of CD8+ T lymphocyte was significantly elevated, which were 27.09±9.86% and 22.08±4.54% respectively (P< 0.05).Conclusion:(1) Comparing to the traditional counting of megakaryocyte, the CV value of standardized counting of megakaryocyte is decreased in patients with ITP and in control group. However there was no apparent difference between the traditional and standardized counting of megakaryocyte. The standardized counting of megakaryocyte maybe decrease the variability compared to the traditional counting of megakaryocyte. Whether it can improve the accuracy of diagnosis to ITP and should be study more.(2) Pathogenesis of ITP involves the disequilibrium of proportion of T lymphocyte subsets and the disorder of their function. Disequilibrium of immune adjustment of T lymphocyte made T-helper (Th) cell insufficient and T-suppresor (Ts) cell excessive relatively, which caused immunity injure of cells. (3) The disorder of T lymphocyte subsets played a critical role in the pathogenesis of ITP. To regulate the balance of T lymphocyte subsets may be a new approach to the treatment of autoimmune disease. |
