Objective1. Analysis the distribution of Arg51Gln and Leu72Met polymorphisms of Ghrelin gene in Chinese Han and Hui nationality in GanSu province;2. To investigate the association with Ghrelin gene polymorphisms and type 2 diabetes mellitus, insulin resistance(IR), blood pressure and lipid in chinese Han and Hui nationality in GanSu province.SubjectsThere were 113 Han patients (58 males and 55 females) with average age of 56.68±13.84 years old, and 106 normal controls (58 males and 48 females) with average age of 55.11±14.98 years old; and there were 137 Hui patients (71 males and 66 females) with average age of 53.80±11.90 years old, and 112 normal controls (68 males and 44 females) with average age of 54.29±14.59 years old;all subjects were unrelated, type 2 diabetic patients were consistent with the diagnostic criteria of WHO,1999, normal control subjects were healthy adults of outpatients of examination at the same period,without serious heart,liver,kidney dieases.MethodsAnthropometrie variables, blood pressure, fasting plasma glucose, fasting insulin,lipid,creatinine,uric acid were measured also. The subjects were genotyped for two single nucleotide polymorphisms (Arg51Gln and Leu72Met) in exon 3 of Ghrelin gene by polymerase chain reaction(PCR)-restriction fragment length polymorphisms(RFLP). Excel was used to build database, SPSS 13.0 was used for analyses.Results1. There were Arg51Gln and Leu72Met polymorphisms of ghrelin gene in chinese Han and Hui nantionanlity in Gansu province.2. There were no significant differences of genotypes distribution of GG,GA,AA,CC,CA,AA and allele fequencies of G, A and C, A of Arg51Gln and Leu72Met of Ghrelin gene between T2DM and NGT both in Han and Hui populations(P>0.05), while there were significant differences of genotypes GG,GA,AA and allele fequencies of G, A of Arg51Gln between Han and Hui nantionnality (P<0.01).3. TC and LDL-C of GA+AA carriers of Arg51 Gin were significantly higher than those with GG in group T2DM in Han and Hui subjects,while TC of GA+AA carriers were significantly higher than those with GG in group NGT (both P<0.05).4. TC of CC of Leu72Met carriers were significatly higher than those with CA+AA in group NGT in Han subjects, TC of CC carriers were significatly higher than those with CA+AA in group T2DM in Hui subjects also,while TC and LDL-C of CC carriers were significatly higher than those with CA+AA in group NGT(both P<0.05).Conclusion1. There were Arg51Gln and Leu72Met polymorphisms of ghrelin gene in chinese Han and Hui populations in Gansu province and there were different genotype frequencies of the Polymorphisms among different races;2. Both Arg51Gln and Leu72Met Polymorphisms weren't related to diabetes;3. The Polymorphisms were association with lipid metabolic disorders.
