Skeletal muscle injuries are common diseases in sports medicine. Skeletal muscle injuries take a long time to recover. After rehabilitation, the original injury is easily to re-injure. The study tried to research the therapeutic effects of Astragaloside and Tanshinone IIA in rat skeletal muscle after acute contusion in the histological and bio-mechanics level, and observe their effects in mRNA level. This study was divided into three parts as following:Objective To study effects of Astragaloside and Tanshinone IIA on the skeletal muscle morphology during the repair process after skeletal muscle contusion.Methods 84 male Wistar rats were divided to six groups:five intervention groups (16 rats in each group)——Astragaloside, Tanshinone IIA, mixed injection (Astragaloside and Tanshinone IIA), Huangqi-Danshen injection and physiological saline, and a normal group (4 rats in this group). In intervention groups, bilateral gastrocnemius of all intervention groups were contused by specific device, then the damaged location was injected with the corresponding drug, and samples were collected from injured muscles after the injury 4,7,14 and 28 days. In the normal group, the bilateral gastrocnemius were collected in the 28 days. To observe the diameter of skeletal muscle fiber (cross section) in HE stain and the area of fibrous scar tissue (longitudinal section) in Masson stain.Results (1) In all groups, the diameters of skeletal muscle fiber had no significant difference at different time. (2) The area of fibrous scar in the 4 medicine intervention groups was smaller than the saline group, the mixed injection group was smaller than astragaloside group and tanshinone IIA group, and the mixed injection group had no difference with the Huangqi-Danshen group.Conclusion During the repair process following acute contusion of skeletal muscle, astragaloside and tanshinone IIA can reduce the area of fibrous scar tissue, and they have no effect on the diameters of skeletal muscle fiber. Objective To study the effects of Astragaloside and Tanshinone IIA on bio-mechanical properties during the repair process after skeletal muscle contusion.Methods The right gastrocnemius of 80 male Wistar rats were contused by specific device. The rats were divided into five groups randomly:Astragaloside, Tanshinone IIA, mixed injection (Astragaloside and Tanshinone IIA), Huangqi-Danshen injection and physiological saline, then the damaged location were injected with the corresponding drug. Intact bilateral gastrocnemius——with a bone at both end of the muscle——were collected after the injury 14,21,35 and 56 days. The failure load and strain rate of the injured gastrocnemius were tested and then compared with those of the normal lateral.Results (1) From the 21 days after injury, the failure load of 4 medicine intervention groups was higher than the saline group. The failure load of the mixed injection group was higher than Astragaloside group and Tanshinone IIA group, and had no difference with the Huangqi-Danshen group. (2) From the 21 days after injury, the strain rate of 4 medicine intervention groups was higher than the saline group. The strain rate of the mixed injection group was higher than Astragaloside group and Tanshinone IIA group, and had no difference with the Huangqi-Danshen group.Conclusion During the repair process following acute contusion of skeletal muscle, astragaloside and tanshinone IIA can improve the failure load and strain rate. But their effects were weaker than the mixed injection and the Huangqi-Danshen injection.Objective To study the effects of Astragaloside and Tanshinone IIA on the expression of MHC-emb and Vimentin mRNA during the repair process after skeletal muscle contusion.Methods The right gastrocnemius of 120 male Wistar rats were contused by specific device. The rats were divided into five groups randomly:Astragaloside, Tanshinone IIA, mixed injection (Astragaloside and Tanshinone IIA), Huangqi-Danshen injection and physiological saline, then the damaged location were injected with the corresponding drug. Samples were collected from injured muscles after the injury 1,4,7,14,28 and 42 days. The mRNA expression of MHC-emb and Vimentin in the muscle was tested by realtime quantitative PCR.Results (1) After the acute contusion of skeletal muscle, the mRNA expression of the MHC-emb and Vimentin in injured muscle in five groups of rats was increased significantly. Respectively reached a peak in the 7-14 days and the 4 days, and then declined. (2) The intervention groups' expression level of MHC-emb mRNA was significantly higher than the saline group. And the expression level of MHC-emb mRNA of mixed injection group and Huangqi-Danshen group were higher than astragaloside group and tanshinone IIA group. (3) The intervention groups' vimentin mRNA expression was significantly lower than the saline group. And the expression level of vimentin mRNA of mixed injection group and Huangqi-Danshen group were lower than astragaloside group and tanshinone IIA group.Conclusion During the repair process following acute contusion of skeletal muscle, astragaloside and tanshinone IIA can promote the expression of MHC-emb mRNA, and inhibit the expression of vimentin mRNA. But their effects were weaker than the mixed injection and the Huangqi-Danshen injection.
