Objective To explore the expression and the significance of minichromosome maintenance protein 2 and nuclei proliferation index Ki-67, and their relationship to cell multiplication in leukemia, In order to provid evidence for pathogenesy, diagnose, treatment and prognosis of leukemia.Methods The experiment is divided to two parts.(1) Leukemic cells K562 and HL-60 are cultivated, and the cell growth curves are observed. The leukemic cells are interfered with Ara-C and their inhibition ratio by MTT is detected. The cells are divided into 5 groups:A group is exponential phase of growth cells.B group is starvation condition cells.C group is the cells intervented by drug of Ara-C.D group is peripheral blood granular cells from health adults.E group is peripheral blood leukomonocytes from health adults. D group and E group are control groups. We detected the express of MCM2 and Ki-67 proteins in each group by immunohistochemistry method. The positive cells are analysised on the way of semiquantitative analysis.(2) Collect 12 primary acute leukemia children as experimental group,and 10 non-malignant hematopathy children as control group, using immunohistochemistry method to explore the expression of MCM2 and Ki-67 in mononuclearcells of bone marrow in each group.Results (1)The expression of MCM2,Ki-67 in exponential phase of growth HL-60 cell and K562 cell is higher than control groups (P<0.05), and the expression of MCM2 is positive correlated with Ki-67 in K562 (r=0.58, P<0.05), in HL-60 (r=0.44, P<0.05).(2) The expression of MCM2,Ki-67 in starvation condition HL-60 cell and K562 cell is higher than control groups (P<0.05) and the groups intervented by Ara-C,but lower than exponential phase of growth cell groups. (3)The expression of MCM2 Ki-67in K562 and HL-60 cells intervented by Ara-C is lower than exponential phase of growth cell groups (P<0.05). (4) The expression of MCM2,Ki-67 in clinical experimental groups is higher than control groups.Conclusions (1) MCM,Ki-67 in exponential phase of growth HL-60 cell and K562 cell is over-expressed. And the expression of MCM2 is positive correlated with Ki-67. (2) Serum starvation and intervention by Ara-C can inhibit cell proliferation and the expression of MCM2 protein,Ki-67 protein. (3) The expression of MCM2,Ki-67 in clinical experimental groups is over-expressed.
