Objective:to develop different PLGA membranes and detect their features, selecte the appropriate PLGA membrane for isolation/scaffold materials, provide experimental evidence for develop the growth factor gene transfer cell-PLGA-elastic membrane.Methods:1.Using DL-PLGA polymers 1-5,which have different LA/GA molar ratio and intrinsic viscosity, to produce PLGA membracesâ… -â…¤on solid substrate,use the solution casting methode in an atmosphere of high humidity environments; 2. Measure degradation time in vitro, hydrophilic, surface angle of PLGA membracesâ… -â…¤; 3.Use SEM to detecte PLGAâ…¢'s surface structure and pore size; 4.Culture mouse embryonic fibroblasts, use inverted microscope to detect the fibroblasts'growth situation on the PLGA membrane; 5.Use MTT method to measure the growth situation of the mouse embryonic fibroblasts on the PLGA membraceâ…¢-â…¤,and draw the growth chart;6. The data were analyzed by SPSS 17.0 System.Results:1.PLGA membracesâ… -â…¤all are transparent with 35 microns thickness, PLGA membracesâ…¢-â…¤'s flexibility is better than PLGA membracesâ… -â…¡'s; 2.he intial degradation time of PLGA membracesâ… -â…¤are about the first week, the second week, the fifth weeks, the tenth weeks and the eighteen-twenty week; 3.LGA membrace â…¢Has the best hydrophilic during PLGA Membracesâ…¢-â…¤,P<0.05; 4. Cells use the shortest time to adhension on membraneâ…¢and have good growth situation; 5.Use SEM to detect membraneâ…¢find there are neatly arranged pore,the miameter is about 3-4 microns.Conclusion:the PLGA membraneâ…¢has good flexibility, it begins to degradation at the fifth week, it has good hydrophilic property and can adhension with fibroblast well, it's surface have good hole diameter, the result meet the experiment requirements, can use it for next exprinment.
