Palmitate Acid With Different Concentration On The Dynamic Effects Of INS-1 Cells' Function, Apoptosis And The Changes Of SREBP-1c Gene

Objective:To observe the changes of insulin secretion, apoptosis, and SREBP-1c gene expression in INS-1 cells on different concentrations palmitate acid.Methods:INS-1 cells were cultured in RPMI1640, the cells were divided into experimental groups and control groups. Experimental groups were treated with different concentration palmitate acid(PA 100μmol/l,200μmol/l,300μmol/l,400μmol/l), the control group were treated without palmitate acid. After 6 hours and 24 hours, the vitality and amount of the cells were estimated by MTT assay; Apoptosis rate were identified by PI staining, then quantitated by flow cytometry; The expression of insulin mRNA,SREBP-1cmRNA and Insig-1mRNA were determined by semi-quantitative RT-PCR.Results:In experimental groups treated with 100μmol/l,200μmol/l, 300μmol/l,400μmol/l palmitate acid for 6 hour, the cells function and apoptosis rate were similar with the control group (p>0.05), basal insulin secretion increased after treated with 100μmol/l,200μmol/l,300μmol/l palmitate acid, but not changed treat with 400μmol/l palmitate acid; the Expression of insulin mRNA was similar in experimental groups and the control group. After 24 hours, the result of MTT assay indicated that vitality of the cells were decreased in experimental groups(p<0.05), apoptosis rate was significant increased, and with the increase of palmitate acid concentration, the basal insulin secretion was reduced, and insulin mRNA expression was downregulated(p<0.05), SREBP-1cmRNA and Insig-1mRNA expression haven't significant different between experimental group and control group(P>0.05).Conclusions:Palmitate acid on INS-1 cells with different length of time leading to different effections, in a short time low concentration palmitate acid promote insulin secretion; INS-1 cell interfered by palmitate acid in a long time will influence the function and promote apoptosis in a dose dependent manner, while SREBP-lc mRNA expression was not significant different.
