Objectives:This experiment applying electron microscopy technology and flow cytometry technology detects impact that the Costasis of hemostatic affects on brain tissue in Sprauge-Dawley rats, and researches the effects of it in department of cerebral surgery.Methods:48 Sprauge-Dawley rats were randomly divided into two groups. One is experiment group (group E, sprayed Costasis on the surface of pallium), and another is contrast group (group C, sprayed physiological saline on the surface of pallium). Each group was divided into four time points which were 1week,2weeks,3weeks,4weeks.6 rats were allocated to each group at these any time points. Then these rats were anesthetized at the time points above-mentioned, and the obtained brain tissues were detected by electron microscopy technology and flow cytometry to observe changes. Another 40 rats were randomly divided into four groups. Each group was 10 rats. Experiment group, to spray CoStasis on the surface of incised pallium. Contrast group 1, to press Gelatin sponge on the surface of incised pallium. Contrast group 2, to sprinkle Arista on the surface of incised pallium. Blank group, nothing was done on the rats. Each rats was made into encephalic bleeding model (The pallium near the right frontal pole was cut by the size of 3mm×2mm×1mm). Then the wounds were dealed with local hemostatics at once, and the complete stopping bleeding time was recorded. At last, the results were analysed with SPSS 17.0 statistical softwares. The P value less than 0.05 was thought to be statistical significance.Results:1 Costasis of hemostatic might have little side effects on the morphosis of brain tissues compared with physiological saline according to observed results during four weeks. The detected results of electron microscopy proved that neuronal structure morphology was normal essentially. Apoptosis morphous, such as nucleolar disappearance, chromosomal close up and so on, hadn't taken place. Flow cytometry also showed that the two groups of apoptosis rates and cellular proportion in Cell Cycle didn't have significant deviation (P) 0.05).2 The average complete stopping bleeding time (ACSBT) of group E was 14.6s, ACSBT of group C1 was 36.7s, ACSBT of group C2 was 25.9s, and ACSBT of blank group was 194.0s. ACSBT of four groups were adopted by LSD-test of multiple comparisons. ACSBT of group E was signifycantly more swift than in group C1 (P<0.05), in group C2(P<0.05) and in blank group (P<0.05).Conclusions:1 Adopting electron microscopy technology and flow cytometry technology confirms that CoStasis has less side effect on the morphosis of rats'brain tissues. 2 CoStasis is superior to Gelatin sponge and Arista with regarding of the effect of stopping bleeding.
