Spinal cord injury, even a partial spinal cord injury may result in permanent partial paralysis which will seriously affect patients'quality of life and bring heavy burden to their families and society. Some researchers found that spinal cord injury can be repaired by cell transplantation, gene therapy or genetically modified cell transplantation. Many research suggest that OECs is the most outstanding cell among all the candidates for cell injury repaired by transplantation in central nervous system.On the cell membrane of OECs there are many moleclues related to cell adhesion and promotion of nerve regeration.They can provide a good internal environment for axonal regeneration and help the growth and elongation of axon. It will help promoting axon extended to the target region which will lead to the recovery of neurological function Many studies has proved that OECs can promote sciatic nerve regeneration and the axons extension in the half-cut model for the growth in mice.Now, NASH differential adhesion and arabinosylcytosin are the most well-established method for culture of olfactory ensheathing cells in vitro.NASH differential adhesion and arabinosylcytosin methods utilizes the different adhension time of fibroblasts, astrocytes and ensheathing cells.OECs can display many cell morphology and cell surface marker when they are cultured. There are usually two kinds of them. One of them is species of neurolemmal cellular type in which the p75NTR receptor will express.They are mainly located at the peripheral olfactory nervous system,in a spindle shape. The other one is species of horizontal cellular type, mainly distributing in mouse olfactory bulb,it is always in a flat shape. They are lack of p75NTR receptor but positive in GFAP. We use GFAP immunohisto-chemical identification to measure the quality of purification and changes between of olfactory ensheathing cells in vitro.In this study, we used a modified differential method of NASH, and Ara-C purification of adherent to isolate and culture the olfactory ensheathing cells from mouse olfactory bulb.The anti-PSA-NCAM and anti-L-NGFr area classification type astrocytes OECs and class Schwann cell OECs, are also used, in order to identificat OECs with different types of expression and secretion. It can be better to graft for repair of spinal cord injury, the promotion of axonal regeneration in the role.
