【Objective】To study the therapeutic effect of through the tail vein of rats injected with large capacity interleukin-10 (IL-10) plasmid DNA induced by pig serum and effects of IL-10 on the expression of tumor necrosis factor-α(TNF-α),platelet derivative growth factor-β(PDGF-β) and cyclooxygenase-2(COX-2)in fibrotic rat liver.【Methods】Thirty SD rats were divided into normal control group(group N),fibrosis model group(group M),rIL-10 gene therapy group(group I)and empty vector control group(group P) randomly. Rats in group N were treated with 0.5ml normal sodium via intraperitoneal injection twice a week for 8 weeks, and rats in other groups were treated with 0.5ml pig serum via intraperitoneal injection twice a week for 8 weeks. At the beginning of the 5 th week, rats in group N and M were injected with Ringer's solution as a reagent control via tail vein weekly, rats in group I were injected with the rIL-10 recombinant plasmid pcDNA3-rIL-10 and rats in group P were injected with empty vector pcDNA3 via tail vein weekly. All rats were sacrificed at the end of 8th week, collecting liver tissue. By HE staining to detect the pathological changes in liver tissue. By SP immunohistochemistry to detect the expression of TNF-α,PDGF-βand COX-2 in rat liver.【Results】The pathohistology analysis confirmed experimental rat hepatic fibrosis model induced by pig serum was successfully constructed. Compared with the rats in group M and P, rats in group I showed rat liver cell degeneration and necrosis was mild, the portal area and inflammatory cell infiltration decreased,collagen deposition decreased. Immunohistochemistry results showed that: compared with the group N , the expression of TNF-α,PDGF-βand COX-2 in group M and group P rat liver was significantly increased, the difference was significant (P <0.01); and compared with group M and group P , the expression of TNF-α,PDGF-βand COX-2 in group I was significantly reduced, the difference was significant (P <0.01).【Conclusions】By intravenous injection of rIL-10 plasmid, rIL-10 gene is highly expressed and the expression of TNF-α,PDGF-βand COX-2 is effectively inhibited in liver tissue.
