| Objective: Through the genetic analysis of eight X-STRs and thirteen X-SNPs with STR genotyping platform and TaqMan technology respectively to provide methods for special cases such as the judgement of relationships of grandmother to granddaughter and half-sib sisters with the same father.Methods: The polymorphisms of eight X-STRs in Chinese Han population were detected by Mentype? Argus X-8 kit. The genetic parameters of them were calculated and applied in practical cases. Thirteen X-SNPs located equilibrately on X chromosome were detected by TaqMan technology. The sensitivity, accuratissime, specificity and adaptivity of samples of TaqMan technology were explored, together with the polymorphism,genetic parameters and linkage information of X-SNPs. In addition, the application value of these genetic markers on X chromosome were investigated.Results: The eight X-STRs included in Mentype? Argus X-8 kit were polymorphic in Chinese Han population with ideal genetic parameters. Loci of DXS10135 and DXS10101 with highest exclusion rate reach 85.51% and 75.36%, respectively. The thirteen X-SNPs which heritaged independently and followed Hardy-Weinberge Equilibrium genotyped by TaqMan technology accurately, with high sensitivity, specificity and adaptivity. The Polymorphic Information Contents (PIC) of them are all above 0.3 and the mean Heterozygosity is 0.4894. Detected the two kinds of genetic markers together, Cumulative Discrimination Power (CDP) in femeles reach 0.999999999999997, in males reach 0.999999998699; Cumulative Probability of Exclusion(CPE) in duos reach 0.999997177, in trios reach 0.999999976392.Conclusion: The eight X-STRs included in Mentype? Argus X-8 kit were suitable for Chinese Han population. The genotyping of X-SNPs by TaqMan technology was accurate and reliable. Thirteen X-SNPs with high polymorphism and independently heritage can meet the demands of forensic application. Statistic analysis show that combined detection of the two kinds of markers can provide considerable genetic information for special cases. |
