Objective:To determine the inhibitory effect of the cell Stellera chamaejasme L.of Blood Serum on the cell proliferation of the human liver cancer SMMC-7721 cell line and the expression of VEGF and Survivin, investigate it in the Molecular Mechanism of the Apoptosis of the Human Hepatoma Cell Line SMMC-7721.Method:the human cervical cancer was cultured by Stellera chamaejasme L.of Blood Serum in vitro. The cells was treated in 24 hour and 48 hour with different concentrations of Stellera chamaejasme L.of Blood Serum and different hour Stellera chamaejasme L.of Blood Serum, end then cell growth was assessed using MTT assay,the change of SMMC-7721 cell was observed by convert light microscope.The technique of flow cytometry was used to measure DNA content. The method of Western Blot was used to measure VEGF and Survivin expressions of changing in 24 hour.Result:MTT test showed that the Stellera chamaejasme L.of Blood Serum could suppress the cell proliferation of the human liver cancer SMMC-7721 cell line. The most remarkable inhibitory effect was the Stellera chamaejasme L.of Blood Serum of two hours collected, 24h inhibited SMMC-721 cell growth was positively correlated, But 48h it was not in dose and time-dependent manner.The increasing necrotic cells had been observed by fluorescent staining, convert light microscope and electron microscope by Stellera chamaejasme L.of Blood Serum in Human Hepatoma Cell Line SMMC-7721. The percentages of Gl/G-phase cells were significantly increased after treatment with Stellera chamaejasme L.of Blood Serum compared with those in control group in 24h, particularly the Stellera chamaejasme L.of Blood Serum of 2h gained were the best. But the effect of 48h less than 24h.The above three experimental results show that the 2 hour Stellera chamaejasme L.of Blood Serum collected had the best efficacy and dose-dependent. Western blotting analysis indicated that the Stellera chamaejasme L.of Blood Serum of various concentrations could reduce the expression of VEGF and Survivin proteins in SMMC-7721 cells. we choosed 5 concentrations of Stellera chamaejasme L.of Blood Serum that those were 3g/kg,6g/kg, 9g/kg,12g/kg and conttrol in Western blotting.Conclusions: the Stellera chamaejasme L.of Blood Serum strongly inhibited cell growth and proliferation of the human liver cancer (SMMC-7721) cells. Particularly, at the concentration of 2h Stellera chamaejasme L.of Blood Serum collected. the Stellera chamaejasme L.of Blood Serum of various concentrations could reduce the expression of VEGF and Survivin proteins in SMMC-7721 cells.
