Human enterovirus 71 is a member of the small RNA virus family, and a member of the genus enterovirus, which is the major pathogen of hand, foot and mouth disease. Hand, foot and mouth disease and central nervous system infection are the common clinical symptoms which are caused by EV71. Currently, no specific and efficient drug is to treat the EV71 infection. Inactived vaccine just entered phase I clinical trial; Recombinant protein vaccine research is still in the animal experiment stage. It is neccessary to look for a kind of effective antigenic protein. The EV71 capsid proteins: rVP1, rVP2, rVP3, rVP4 are expressed efficiently in the E.coli in this research, the expressed products are the form of inclusion bodies. The SDS-PAGE purity is above 90% after inclusion bodies are purified and refolded. It is shown in the amimal experiments that rVP1, rVP4 protein are good in the immunogenicity, and rVP2, rVP3 proteins are no. Only rVP1 protein can induce the neutralizing antibodies. Recombinant VP4 protein is a not potential recombinant protein vaccine withourt inducing the neutralizing antibodies. At the same time, rVP2,rVP3 expressed in the E.coli are different from the EV71 virus particle surface VP2, VP3 proteins in the conformation. The further study is needed on the VP2, VP3 proteins. In this paper it is shown that VP1 is a potential candidate antigen for the recombinant protein vaccine. It provides an important basis for screening the EV71 vaccine candidate antigens.
