Objective To investigate different gene expression of DCs responses to Human alloantigens and Cytomegalovirus Antigen, we want to find specific gene expression programs or specific signal pathway of DC responses to alloantigens which will be target for preventing or treating GVHD.Methods We use Microarray technology to investigate different gene expression of DCs responses to alloantigens and Cytomegalovirus Antigen. Modulated genes were selected and clustered on the bases of KEGG pathways and sorted according to their biological function using a P value cutoff of 0.05.Result1. At 12h, Cytomegalovirus Antigen induced modulation of 3,335 mRNA species, among these 1608 induced at least 2 fold,but alloantigens only induced modulation of 122 mRNA species, among these 7 induced at least 2 fold. At 24h, Cytomegalovirus Antigen induced modulation of 2,386 mRNA species, among these 1053 induced at least 2 fold ,alloantigens induced modulation of 442 mRNA species, among these 31induced at least 2 fold.2.These common signaling pathway involved in antigen processing and presentation , toll-like receptor signaling pathway and cell movement/ migration, et al. Metabolism of xenobiotics by cytochrome P450 is specific signaling pathways for All-DC Those specific signaling pathways involved in proteasome, ubiquitin mediated proteolysis,SNARE interactions in vesicular transport for CMV-DC. Conclusion1. Human alloantigens can induce DCs genes moderate modulation;2. The gene expression of DCs responses to human alloantigens and Cytomegalovirus antigen are much different;3.We selected 16 differentially modulated genes, previous results indicated that among these HSP70,HSP90,TNFSF14,CSF1 have the potent for preventing or treating GVHD ,but others need to future investigate;...
