Objective To investigate the effects of human mesenchymal stem cells (hMSCs) on proliferation and apoptosis of pancreatic tumor cell line Aspc-1, exploring the role of hMSCs in stroma of solid tumor and providing theoretical basis for use of hMSCs in treatment of pancreatic cancer.Methods hMSCs was isolated from adult bone marrow by density-gradient centrifugation and cultured with low-glucose culture media. The surface markers were identified by immunohistology. Conditioned media of hMSCs was put into culture system of Aspc-1. Proliferation and apoptosis of Aspc-1 were individually evaluated by MTT method and flow cytometry. Expressions of GSK-3βand Bcl-2 in Aspc-1 were measured by immunohistology and western blotting.Results With the influence of conditioned media of hMSCs , growth inhibition rate and apoptosis rate of Aspc-1 were increased. Meanwhile, expression of GSK-3βin Aspc-1 was increased and expression of Bcl-2 in Aspc-1 was decreased. All results were statistically significant(P<0.05).Conclusion hMSCs can inhibit the proliferation of Aspc-1 and enhance its apoptosis . hMSCs inhibits the proliferation of Aspc-1 probably by upregulation of GSK-3βin Aspc-1, and enhances its apoptosis by downregulation of Bcl-2 in Aspc-1.
