Objective:To investigate the effect of plumbagin on proliferation and apoptosis of human melanoma cell, A375 in vitro.Methods:Various concentrations of plumbagin were used to treat A375 cell for 24h. Cell proliferation was determined by MTT assay. Cell apoptosis-inducing effects were determined by using flow cytometry with Annexin V-FITC and PI double labeling method. The expression of NF-κBp65 and bcl-2 at protein levels were detected by western blot.Results : Compared with the blank control group, plumbagin(1 -13μmol/L) could significantly inhibit the proliferation in a dose-dependent manner(p<0.05). After 24-hour treatment, the apoptosis rate was 8.52 %±0.96%,14.83%±1.34%,19.56%±1.85% in A375 cells treated with plumbagin of 2.5,5.0,10.0μmol/L, respectively, compared to 4.58%±0.46% in untreated cells (p<0.05). The expressions of NF-κBp65 and bcl-2 at protein levels were down-regulated in plumbagin-treated groups (2.5,5.0,10μmol/L) compared with blank control group(p<0.05).Conclusion:Plumbagin could induce cell apoptosis and down-regulate the levers of NF-κBp65 and bcl-2, as well as inhibit the proliferation of melanoma A375 cells in vitro.
