Methods The SD rats bone marrow mesenchymal stem cells were isolated and cultured in vitro. The third passage cells were harvested and exposed with EMF at 15Hz 1mT for 8h/d. The semi-quantitative RT-PCR technique was used to measure PTHrp,Agc I,II collagen and Sox9 mRNA expressions; Western blot was used to measure the type II collagen expression. After the inhibitor of protein kinase A (PKA ) H-89 or the inhibitor of protein kinase C (PKC) Go-6976(12μm) were added, the effects of EMFs on Agc I,II collagen and Sox9 mRNA expressions were measured again by RT-PCR;and the II collagen protein expression was detected again by Western blot. Results EMFs of 15Hz 1mT induced significant increase of mRNA expressions of PTHrp,Agc I,II collagen and Sox9 in comparison to the controls;and promoted the type II collagen protein expression. The Agc I,II collagen expression reduced when PKA pathway inhibitor H-89 or PKC inhibitor Go-6976 were added, but the mRNA expression of Sox9 was not affected.Conclusion EMFs at 15Hz 1mT can promote the markers of chondrogenesis differentiation expressions of bone marrow mesenchymal stem cells. The effect is correlation with PKA and PKC pathways.
