The Protective Effect Of Remote Ischemic Preconditioning On Rat Muscle Flap Against Ischemic Reperfusion Injury

Objective To observe and compare the protection of the remote ischemic preconditioning (remote IPC) and classic ischemic preconditioning (classic IPC) on rat muscle flap against ischemic reperfusion injury, and detecting the SOD in rat blood serum and muscle tissue to descript the SOD make a important role in remote IP.Methods 48 SD rats divided into three groups by the different experiment operation randomly. In the group A (control group), a 3-hour flap ischemia was induced after preparation of the latissimus dorsi muscle; in group B (classic IPC group), a two times of 10 min brief flap ischemic induced by preclamping the pedicle and then by 30 min reperfusion; in group C (remote IP group), the limb artery was clamped by two times of 10 min. After 30 min reperfusion, the group B and group C flap suffered 3-hour ischemic as group A. Each group divided into two sub groups randomly. The blood and muscle tissue harvested in each sub group at different time to detect the SOD and MDA. The muscle flap harvested in 6 hours after reperfusion and 5 days after operation in each sub group. Results Two experiment groups decreased the necrosis area compared with control group (P<0.01). Under microscope, the degeneration of muscle tissue in experiment group lessened than control group. Two experiment groups had a higher SOD and lower MDA in blood serum at each point than control group. 3-hour after ischemia and 1, 6 hour after reperfusion, the muscle tissue of two experiment groups had a higher SOD and lower MDA at each point than control group. But at the 3-hour after ischemia and 1 hour after reperfusion, group 1 changed obviously than group 2 in muscle tissue.Conclusions Remote IP has a good protection on rat muscle flap against ischemic reperfusion injury the same as Classic IP. The mechanism of the protection may associate the activity of SOD.
