Objective: To investigate the effect of 5-Aza-CdR on the re-expression of CDH13 and the function of it on human breast cancer cell line MCF-7.Methods: The breast cancer cell line MCF-7 was treated with different concentration of 5-Aza-CdR.The effect of 5-Aza-CdR on proliferation was evaluated by MTT assay. The cell cycle was analyzed by flow cytometry. RT-PCR were used to determine the expression of CDH13 in the MCF-7 cell after the treatment of 5-Aza-CdR.Results:The cell growth speed was significantly slowed down and the cell cycle was inhibited at G0/G1 phase after the treatment of 5-Aza-CdR. The flow cytometry did not detect the apoptosis .5-Aza-CdR can also restore the expression of CDH13mRNA in MCF-7 cells. Conclusion: CDH13mRNA was silenced by aberrant promoter region methylation in MCF-7 cells. 5-Aza-CdR can restored the expression of CDH13mRNA and inhibited cell proliferation, which suggested that the restoration of CDH13 expression may be one of the mechanisms for the biological behaviour changes of MCF-7 cell.
