Purpose: To study the chemical constituents of gorgonian Astrogorgia dumbea and soft coral Dendronephthya sp. collected from the Dongshan island of East China Sea, and research the cytotoxic activitiy of some compounds isolated from gorgonian A. dumbea.Method: The chemical constituents of A. dumbea and D. sp.were obtained by the methods of extraction, recrystallization, column chromatography and semi-preparative HPLC, etc. Their structures were elucidated by the spectroscopic methods (1D, 2D-NMR; EI-MS, HR-ESI-MS; IR, UV, etc.).Result: Twelve compounds were isolated from the MeOH and CHCl3 extractions of A. dumbea. And the structures of nine compounds (see Fig. 1) were identified as dimorphoside A (LAD-01), astrogorgiaside A (LAD-03), astrogorgiaside B (LAD-04), astrogorgiaside C (LAD-05), astrogorgiaside D (LAD-07), astrogorgiaside E (LAD-09), astrogorgiaside F (LAD-10), astrogorgiaside G (LAD-11); and batilol (LAD-12). The others were still under indentifying.Four compounds were isolated from the n-BuOH and CHCl3 extractions of soft coral D. sp.. Two of them were identified as pregna-1,20-dien-3-one (LSC-04) and pregna-1,4,20- triene-3-one (LSC-06), respectively (see Fig 2). The others were still under indentifying.The cytotoxic activity of some compounds isolated from gorgonian A. dumbea was tested. All tested compounds have cytotoxic activity against Bel-7402. At the same time, dimorphoside A and astrogorgiaside A have activity against HL-60; and astrogorgiasides B and C have cytotoxic activity against K562.Conclusion: Most of compounds from A. dumbea were steroid saponins which astrogorgiasides A-G of them are novel compounds. Dimorphoside A and batilol were first isolated from Astrogorgia species. Two pregnane compounds from D. sp., LSC-04,LSC-06, were isolated from this species firstly.
