ObjectiveSIRT1, SIRT4 have been confirmed to be associated with homeostasis of glucose/lipidmetabolism, but their roles in human type 2 diabetes (T2DM) are poorly understood. This studywas designed to investigate the relationship between SIRT1, SIRT4 in peripheral bloodleukocytes (PBLs) and T2DM.MethodsPeripheral blood and biochemical data were collected from 52 healthy individuals (thenormal control group, NC group) and 113 cases of T2DM patients. Immunocytochemicalstaining was used to detect SIRT1, SIRT4 and reverse transcription polymerase chain reaction(RT-PCR) was used to detect SIRT1 or SIRT4 mRNA levels in PBLs. All data were analyzed bystatistical software application SPSS 17.0.Results1. Immunocytochemical staining showed that SIRT1 was expressed in both nucleus andcytoplasm, and SIRT4 in the cytoplasm of granulocytes and monocytes. No SIRT1 or SIRT4was found in lymphocytes.2. RT-PCR showed that, SIRT1, SIRT4 mRNA levels in T2DM group were lower than thosein NC group (P <0.01).3. Correlation analysis showed that there was a negative correlation between SIRT1, SIRT4and fasting plasma glucose (FPG) (P <0.05)(r= -0.161 and -0.156), a negative correlationbetween SIRT4 mRNA levels and triglyceride(TG)/lipoprotein a (LPa) levels (P <0.05), and apositive correlation between SIRT4 mRNA levels and high density lipoprotein cholesterol(HDL)(P<0.05).4. Fasting plasma glucose (FPG), triglyceride (TG), lipoprotein a (LPa) of T2DM group weresignificantly higher than those of NC group (P <0.01). The body mass index (BMI), systolicblood pressure (SBP) and diastolic blood pressure (DBP) of T2DM group were significantlyhigher than those of NC group (P <0. 01).Conclusion1. SIRT1, SIRT4 were expressed in PBLs (granulocytes, monocytes). 2. SIRT1, SIRT4 may have a role in the pathogenesis of type 2 diabetes (T2DM); SIRT1,SIRT4 in PBLs may indirectly reflect the homeostasis of glucose/lipid metabolism andpancreatic isletβ-cell in T2DM.
