| Purpose:Make the research about the Anti-cancer effect and mechanism of the prescription of Xiaochaihutang combined cyclophosphamide affect EAC hepatumor cells in mice.Material and method:Taking 40 Kunming mice.All the mice were injected EAC hepatumor cells(2.0×10~6/ml,0.2 ml/single)under the left armpit subcutaneous in sterile conditions. After 24 hours were randomly divided into simple tumor group,Traditional Chinese medicine group, western medicine group, the joint group.Then we used the prescription of Xiaochaihutang,cyclophosphamide,the prescription of Xiaochaihutang combined cyclophosphamide to deal with the mice respectively.After ten days, the mice were killed, stripping tumors and calculate inhibitory percentage of tumor cell;tumor cell morphology change were observed by HE dyeing;the ultrastructure of tumor were observed under transmission electron microscopy;observed the expression of Bcl-2 protein by immunohistochemistry method.Results:1.The general state of mice in each group: the simple tumor group has gloomy color fur,sport and mental condition was very poor;TCM group has slightly color fur, the movement was smooth,the mental state was better; western medicine group has a little gray color fur, sport and mental condition was poorer,some mice appeared loose stools and nasal bleeding; joint group mouses have bright and smooth fur,sport flexible,mental condition was very good.2.After drug treatment,compared with the simple tumor group,the tumor of every treatment group were different degree of heavy reduce.The inhibitory percentage of tumor cell was 33.62%(Chinese traditional medicine group), 31.59%(western medicine group),38.85%(joint group).3.Observed the tumor cells under light microscopy, simple tumor group can be seen tumor cell growth exuberant, bigger tumor cells, the nucleus increases and deeply stained, also has obvious pathological karyokinesis. Traditional Chinese medicine group, western medicine group and joint group of tumor cell growth inhibition, the size of tumor cell decreased, nuclear become pyknotic and pathological karyokinesis were less than simple tumor group. The joint group tumor cells arranged in a rule with minimum atypia,tumor cells differentiation was best,pathological karyokinesis was less.4.By electron microscopy observation tumor cells,simple tumor group tumor cells were in a round or irregular shape, the mitochondria and endoplasmic reticulum were expansion in the cytoplasm, the nucleus was larger and clearer, nucleoli was obvious.The size of tumor cells were obvious decreased in Traditional Chinese medicine group, western medicine group and joint group.nuclear structure were anomalous,nuclear chromatin become pyknotic, Typical apoptotic bodies can be seen in tumor sells.the amount of apoptotic bodies which have been seen in joint group was more than other groups.5.Immunohistochemistry showed,traditional chinese medicine group,western medicine group and joint group Bcl-2 were positive expression, gray values were lower than the simple tumor group,joint group to reduce the most obvious.Conclusion:1.The prescription of Xiaochaihutang combined cyclophosphamide could enhance the quality of life of the EAC liver cancer mice.2.The prescription of Xiaochaihutang combined cyclophosphamide can inhibit EAC liver tumor cell's growth.3.The prescription of Xiaochaihutang combined cyclophosphamide have the function of promoting tumor cell apoptosis.4.The prescription of Xiaochaihutang combined cyclophosphamide may reduce liver cancer in mice EAC tumor Bcl-2 protein expression,then achieve inhibiting tumor role.5.The prescription of Xiaochaihutang combined cyclophosphamide can improve the EAC liver cancer mouse survival quality,suppresses the function of tumor grows,which is through the induction tumour cell perishes weakly, the promotion TNF-αproduces the function mechanism realizes.6.The prescription of Xiaochaihutang combined cyclophosphamide can reduce side effects and low toxicity, increase the role of drug efficacy.
