ObjectiveTo investigate the effect of DATS on CLP-induced acute lung injury(ALI) in rat.MethodsIn this experiment, male SD rats were studied, 30 male SD rats were fasted 16 hours before surgery, anesthetized by intraperitoneal injection of 10% chloral hydrate (350mg/kg), and than divided into 3 groups randomly(n=10 per group): control group, CLP group and CLP+DATS group. Control group ware exposed intestine after the laparotomy, and then layered suture off the abdominal cavity, the rat received intravenous injection of normal saline 10ml/kg after the surgery; CLP group ware made into acute lung injury by the cecal ligation and puncture; CLP+DATS group were intravenously injected DATS 10mg/kg after the surgery. All SD rats were injected with normal saline 5ml/100mg by Subcutaneous injection, the rats were killed 12 hours by the blood and collected samples of lung tissue, measured the lung tissue wet/dry weight ratio (W/D), myeloperoxidase (MPO) activity, malondialdehyde (MDA), superoxide dismutase (SOD) and nitric oxide (NO) levels, and observed under light microscope for pathological changes of lung tissue. All data are mean±standard deviation ((x|-)±s) that use SPSS11.5 for windows statistical software for data analysis, groups were analyzed using ANOVA, P <0.05 for the difference was statistically significant. ResultsAlveolar changes in lung tissue in Control rat was not obvious under the light microscope without edema and inflammatory cell infiltration; CLP Group ware observed that the alveolar wall was alveolar wall thicken and edema with inflammatory cell infiltration, destruction of alveolar structure, focal atelectasis and exudates in the alveolar. and the pathological changes in CLP+DATS were significantly reduced compared with CLP group (P <0.05), However, higher than that in control group. Compared with the control group, Lung W/D, MPO activity, MDA and NO levels increased significantly, SOD activity was decreased (P <0.01) in CLP group, but CLP+DATS groups compared with the CLP, Lung W/D, MPO activity, MDA and NO were decreased, SOD activity increased (P <0.05).ConclusionDATS can reduce the CLP-induced lung edema, inflammatory cell infiltration, reduced MPO activity, MDA and NO levels, increased SOD activity, and it may have a protective effect on acute lung injury.
