Objective: The effects of purarin on the cell proliferation and hepatic differentiation of D3 Mouse Embryonic stem cells were investigated. Methods: MTT was applied to investigated the proliferation of embryonic stem cells. FACs (Flow Cytometry) used to detect hepatocytes together with RT-PCR used to detect expression of hepatic marker at mRNA level, were applied to study the function of purarin in hepatic differentiation from D3-mES.Results: D3-mES could differentiate into cardiomyocytes, smooth muscular myocytes, endothelial cells and hepatocytes. Purarin enhanced the cell proliferation of D3-mES. This effects began at 0.1μM and maximum effects came at 10μM. 100μM increased the cell number of AFP and albumin postive cells, as well to increased the expression of AFP and albumin at mRNA level. Transcriptors BMP4 and BMP2 were exrepssed at higher level after treated by purarin.Conclusions: Purarin enhance the cell proliferation and increased the hepatice differentiation of D3-mES via transcritors of BMP4 and BMP2.
