Affiliated Union Hospital of Tongji Medical College Huazhong University of Science and Technology Master degree candidate: Zhao Zhixiang Supervisor: Prof. Tao ShaotangObjective:To understand the relation between biliary atresia (BA) and regulatory T cells (Treg cells) , the studies were performed on the ratio of Treg cells to total blood T cells and the positive staning in the liver with FoxP3 of biliary atresia patients , congenital dilatation of the bile duct and healthy controls. Methods:Each 2 ml venous blood of 86 patients (18 cases are biliary atresia group, 29 cases are congenital cystic dilatation of the bile duct and 39 cases are normal control group) was examined on the ratio of CD4+CD25+T cells to CD4+T cells,CD4+CD25highT cells to CD4+T cells,CD4+CD25highFoxp3+T cells to CD4+T cells by flow cytometry. To gather venous blood and detect the serum level of hepatic function index: ALT by auto-biochemical analyzer. The liver specimens were obtained from the 86 patients. The expression of CD4+ cells,CD8+ cells and Foxp3+ was determined from 4% paraformaldehyde-fixed, paraffin-embedded tissues of livers with innumohistochemical technique.Results:The both ratios of CD4+CD25highT cells to CD4+T cells of biliary atresia and congenital cystic dilatation of the bile duct group were lower significantly than the average control group (P <0.05; P <0.01). The ratio of CD4+CD25highFoxP3+T cells to CD4+T cells of biliary atresia group and congenital cystic dilatation of the bile duct group was separately lower significantly than the normal control group (P<0.01;P <0.05). The both ratios of CD4+CD25highT cells to CD4+T cells and CD4+CD25highFoxP3+T cells to CD4+T cells were nosignificant different between biliary atresia group and congenital cystic dilatation of the bile duct group.There was significant negative correlation(r=-0.722, p=0.043<0.05) between ALT and ratio of CD4+CD25highFoxP3+T cells to CD4+CD25highT cells in the patients whose quantitative value of ALT was not below 80U/L in biliary atresia group.In the biliary atresia group, the positive staning with CD4+ cells and CD8+ cells were higher significantly than control group; the positive staning with FoxP3 was lower significantly than control group. Conclusion:It is showed that the regulatory T cells in children with biliary atresia are defect and regulatory T cells it may be one of the important risk factors on biliary atresia.
