| Objective:The research is to observing vascular endothelial growth factor(VEGF) in blood serum after animal model of AS interfering by simvastatin, exploring the effect of simvastatin toward to the expression of VEGF. Evaluate the effect of VEGF toward to development process of atherosclerotic plaque and vulnerability of atherosclerotic plaque by observing early term pathology results of atherosclerotic plaque interfering by simvastatin or not.Methods:18 New Zealand white rabbits were divided into three groups randomly, group A(normal control), group B(model of AS), group C(simvastatin intervention).Group A and B were fed a 2% cholesterol diet for two weeks. The rabbits'right femoral artery was surgically exposed, a small arteriotomy site created, and a 4F Fogarty embolectomy catheter was advanced into the abdominal aorta up to the level of the diaphragm. The catheter was then inflated to a pressure and pulled to the bifurcation of the aorta. After 3 such passes, the surgical cutdown site was closed.The animals were continued on the hyperlipidemic diet for 12 more weeks and were made the atherosclerotic model. Group C were interfering by simvastatin. The rabbits were euthanized.The entire length of the aorta was exposed and was divided several segments.Then histological assessment of experimental atherosclerosis, plasma VEGF level was measured by enzyme linked immunoserbent assay (ELISA). At last, statistical analysis was made.Results:1. VEGF level was decreased in group C interfering by simvastatin than in group B;2. Cap / core ratio of atherosclerotic plaque was increased in group C interfering by simvastatin than in group B;3. Linear correlation analysis show that VEGF level in blood serum with increasion of cap / core ratio of atherosclerotic plaque has negative correlation.Conclusion:Serum VEGF plays a critical role in the development of vulnerability of atherosclerotic plaque, and has the index role in vulnerability of atherosclerotic plaque.
