Sample preparation is essential to bioanalysis. As the speed of analysis going faster, sample preparation has become the bottleneck in bioanalysis. As a result, it's of importance to develop a simple, fast and efficient sample preparation technology in order to improve the quality of the sample analysis. This paper intends to improve some shortcomings in the sample preparation process.There are two major sample preparation methods, i.e. solid-phase extraction (SPE) and liquid-liquid extraction (LLE). Solid-phase extraction can get purer samples, but it costs more and is time-consuming; Liquid-liquid extraction still plays an important role in sample preparation. However, the traditional liquid-liquid extraction operation requires the use of a separation funnel and is tedious and complicated. At the same time, the traditional LLE method is also time-consuming and prone to interfacial emulsification. The study intends to use the special porous material as solid support, which can replace the use of a separation funnel, to establish a new sample preparation technique which can greatly simplify the work and improve the efficiency.This paper determines the appropriate porous material and the special treatment of the media based on a series tests. Evaluate the new sample preparation technique using samples with different properties and compared with traditional method. Result shows that the improved method is low-cost, easy to use and suitable for trace sample analysis.; in addition, it is suited for the use in a 96-well plate system which can provide high-throughput quantitative analysis.We applied the new technique to practice, such as measurement of pesticide carbofuran residues in water, chlorpheniramine maleate content in serum and the content of preservatives in drink The results show that the new technique can play a great part in the field of environment, food and biological analysis.In this paper, we also investigate rapid determination of melamine in milk. Quantitative detection of melamine now mainly rely on HPLC, GC-MS, LC-MS/MS, which have the weakness of high cost, long submission cycle and difficult to fast promotion. We establish an rapid measurement of melamine with composite SPE column combined with UV. After repeat experiment of the structure of composite SPE column, eluent and elution, we initially establish a relatively complete sample preparation system. In addition, the method is verified accurate and feasible through a series of methodologies.
