| OBJECTIVE: To study the expression ofΔNp73,p53,p21waf-1 in invasive ductal carcinoma of the breast,and discuss the relativity among them and investigate their relationship between the expression and 5-year survival rate and clinical pathological characteristics.METHODS: From January 1999 to December 2003,79 cases of breast invasive ductal carcinoma with complete clinical and pathological materials were selected in First Affiliated Hospital of Henan University of Science and Technology Department of Pathology.Protein expression ofΔNp73,p53 and p21waf-1 were examined in these cases by immunohisto-chemical mathod. Data of the expression ofΔNp73,p53,p21waf-1 in breast carcinoma and the relationship between the expression and 5-year survival rate and clinical pathological characteristics ,were analyzed by SPSS16.00.RESULTS:1. The expression ofΔNp73 in invasive ductal carcinoma of the breast was much more than that in normal breast and breast benigh tumor, the difference has the significance statistically; The protein expression level ofΔNp73 was correlated with axillary lymph node metastasis and 5 year survival rate, it was not correlated with age, the site of breast carcinoma and histological grade.2. The expression of p53 in breast carcinoma was much more than that in normal breast and breast benigh tumor, the difference has the significance statistically; The protein expression level of p53 was correlated with histological grade and axillary lymph node metastasis and 5-year survival rate,it was not correlated with age and the site of breast carcinoma.3. The expression of p21 in breast carcinoma was less than that in normal breast and breast benigh tumor, the difference has the significance statistically; The protein expression level of p21 was not correlated with age, the site of breast carcinoma, histological grade, axillary lymph node metastasis and 5 year survival rate.4. The expression ofΔNp73 in breast carcinoma was positively correlated with the expression of p53,CD44V6;The expression of p21 was not correlated with the expression ofΔNp73,p53.5. Univariate factor analyses showed the factors affecting overall breast invasive ductal carcinoma survival were age, axillary lymph node metastasis status,ΔNp73 protein, P53 protein, CD44V6 protein expression; not affecting the overall survival factors were histological grade, P21 protein expression.6. Multivariate factor prognostic analyses of Cox proportional hazard model showed that the expression level ofΔNp73 and lymph node metastasizing or not are the prognostic separate factors. impacting on invasive ductal carcinoma of the breast.CONCLUSION:1.ΔNp73 plays a role in breast carcinoma, breast carcinoma patients who expressΔNp73 protein are more likely to develop lymph node metastasis and the 5-year survival rate decreases.2. As p53 is involved in the process developing breast carcinoma, P53 positive patients are likely to develop lymph node metastasis and 5-year survival rate decreases.3. p21 is probably related to the formation of breast carcinoma, but the progress of breast carcinoma and the incidence of late stage have little to do.4.ΔNp73 and p53 is likely to play the similar roles in breast carcinoma,ΔNp73 and CD44V6 is likely to play the cooperated roles in the mechanism for lymph node metastasis of breast carcinoma, and affects the prognosis of the patients. p21 involved in the formation of breast carcinoma may be the way to play the role through the non-functional p53.5. Age, axillary lymph node status,ΔNp73 protein, P53 protein, CD44V6 protein expressions influence survival rate of breast carcinoma; histological grade, but P21 protein does not.6.ΔNp73 expression and the prediction of lymph node metastasizing or not can serve as independent prognostic indicator of breast carcinoma. |
