Objective: To evaluate the effects of simvastatin on young rats bone formation osteogenic relative genes of proximal tibia and on osteogenic differentiation of bone marrow stromal cells(BMSCs).Methods: Either simvastatin and vehicle administered subcutaneously to the cervical region of the back of sixty SD rats. At the 1st,2nd,3rd week, the rats was sacrificed. We marked them D1,D2,D3,S1,S2,S3. The BMP-2,MMP-13,VEGF of the proximal tibia trabecular bone was analysis by immunohistochemical staining. BMSCs isolation, cultivation, and differentiation in vitro were derived from the femur. The BMSCs were for osteogenic differentiation cultured. ALP staining was performed on the 2nd week and von Kossa staining on the 4th week. During osteogenic differentiation in vitro, Real-time reverse-transcriptase polymerase chain reaction (PCR) analysis was used to investigate gene expression level of mRNA about BMP-2,RUNX2,Osterix,MSX2,DLX3,DLX5 on the 3rd week.Ressults: (1) Immunohistochemical staining: the expression of BMP-2,MMP-13,VEGF in the simvastatin groups and vehicle groups were no significient difference. (2) The ALP positive-stained cells percentage were about the 30% in all the groups (D1:0.29±0.13 , S1:0.32±0.10 ,D1:0.28±0.15 , S1:0.30±0.13 , D1:0.31±0.13 , S1:0.29±0.14)(P>0.05).(3)According to the Real-time PCR analysis, mRNA expressions of the BMP-2,RUNX2,Osterix,MSX2,DLX3,DLX5 in simvastatin groups shown no significent difference compared to vehhicle groups (P>0.05).(4)No different biomineralization found in simvastatin and vehicle groups was detected by von Kossa stained (P>0.05). Conclusions: (1)Simvastatin could not affect the BMP-2,MMP-13,VEGF expressions of bone formation. (2)Simvastatin could not change the osteogenic differentiation and BMP-2,RUNX2,Osterix,MSX2,DLX3,DLX5 mRNA expression in osteogenic differentiation cultured rat BMSCs.
