There was a special class of neurodevelopmental toxicity on studyingorganophosphorus pesticide chlorpyrifos. It could lead to subtle change innerve cells. But so far, it was few study from the perspective of cell polarity.Par polarity complex was a class of proteins that could regulated cellpolarity. In this paper, we study the effects of low concentration ofchlorpyrifos on the development of embryonic neural cell polaritycomplex.By constructing model of pregnant mouse ,pregnant mouse wereinjected 5mg/kg of chlorpyrifos in mouse embryonic neural development ofE7.5-E11.5.In the E12,E14 and E16,pregnant mouse were sacrificed .Inorder to annlysis the effects of neural cell polarity and polarity complexafter exposing low concentrations of chlorpyrifos, it could use somemethods such as the detection of cell morphology of embryo lateral VZ, measurement of cell stereology and the expression of polarity protein Par6and aPKC, the results were as follows:(1) It was no effect on the morphology of embryo and embryo survivalafter exposing low concentrations of chlorpyrifos. But, chlorpyrifos couldpromote the increase of body weight and head circumference in thedevelopment of mouse embryonic.(2) By DiI staining, It could be seen that ventricular radial glia gapbecome bigger and the cells irregularly arranged, cell polarity disruptedafter the exposure to the chlorpyrifos. And from the stereological analysiswe found that chlorpyrifos may promote cell surface area and increasevolume density. All of these indicated the chlorpyrifos would promote cellswelling(3) Immunofluorescence annlysis showed that the expression of Par6 anaPKC could be affected after exposing chlorpyrifos, which could decreasethe expression of Par6 and aPKC in E12 and E16.The expression of Par6was not significantly changed, but chlorpyrifos could increase theexpression of aPKCThe results showed that cell polarity of VZ cells were disordered andthe expression of Par6 and aPKC would change after exposing lowconcentrations of Chlorpyrifos in the early embryo.
