【Objective】To study the therapeutic effects and mechanism of total caffeoyl quinic acid ester on IgA nephropathy in rats【Methods】(1) 44 healthy male Sprague Dawley rats were fed with normal diet for one week and then 24 urine protein and urinary erythrocyte count were measured.(2) These rats were randomly divided into two groups: normal control group (n=13) (Group A) and model group (n=31). IgA nephropathy model was induced by immunization of bovine serum albumin, staphylococcal enterotoxin B ,carbon tetrachloride and freund's adjuvant for 10 weeks. The success of the model was confirmed by urinary erythrocyte counts, 24-hour urine protein, serum creatinine, blood urea nitrogen and IgA immunofluorescence in 3 randomly chosen rats from these 2 groups respectively. The IgAN model rats were randomly divided into two subgroups: the model control group (n=9) (Group B) and the total caffeoyl quinic acid ester-treated group (n=14) (Group C). The total caffeoyl quinic acid ester was administered intraperitoneally at doses of 3 mg/kg/day in Group C for a period of 8 weeks. Urinary erythrocyte count(UEC),urineβ2 Microglobulin ,24-hrs urinary protein excretion , serum creatinine, blood urea nitrogen were measured at the 10th and 18th weeks respectively and the rats were executed. The deposition of immunocomplex in the renal tissue and the degree of mesangial matrix hyperplasia in the mesangial region were measured. The level of expression of gene and the protein of CD71 and TGF-β1 in the renal tissue were determined by immune-histochemistry and the reverse transcription-polymerase chain reaction (RT-PCR) method and a semi-quantitative analysis was conducted. 【Results】(1)The urinary protein quantitation and urinary erythrocyte was negative. prior to experiment. (2)The 24-hour urine protein and urinary erythrocyte count were different significantly in the model group comparing with those of the normal contrast group(P<0.05 in all cases), while the urineβ2-microglobulin and systolic blood pressure had no significant difference in the model group comparing with those of the normal contrast group (P>0.05 in all cases).(3) The difference in a comparison of urinary protein count, urinary erythrocyte count,the serum creatinine and the blood urea nitrogen of the model group with those of the normal contrast group is of statistical significance (P<0.05), that with those of the model group being also of statistical significance (P<0.05). (4) Immuohistochemistry and the measurement of CD71 content by RT-PCR show that the content of CD71 and TGF-β1 in the renal tissue of SD rats had a rise which is an obvious statistical disparity (P<0.05) as compared with the model group.【Conclusions】An examination of the urine of healthy SD rats indicates that 24-hour urine protein and urinary erythrocyte count are negative. The IgAN model of SD rats'were successful by means of bovine serum albumin +freund's adjuvant +staphylococcal enterotoxin B+ carbon tetrachloride. Total caffeoyl quinic acid ester was administered to make a notable improvement of the urine protein and urinary erythrocyte count of the victims as an intended model, to lower their blood urea nitrogen and serum creatinine and to reduce the deposition of immunocomplex in the renal tissue. Total caffeoyl quinic acid ester can effectively inhibit the demonstration of CD71 and TGF-β1 in the renal tissue at both the immunohistochemistry level and the mRAN level. This may be one of the mechanisms wherewith to cure IgA renal disease.
