The Therapeutic Mechanism Study Of Acupuncture And Nerve Growth Factor On Cerebral Palsy

Objective:By research the influence of acupuncture and nerve growth factor therapy on hypoxic-ischemic brain damage (HIBD) rats'overall function, apoptosis and anti-apoptotic gene expression and brain tissue pathological , to study the therapeutic mechanism of acupuncture and nerve growth factor on cerebral palsy and provide theoretical evidence for clinical treatment on the patients with CP and sequelae of CP.Research contents and methods:1. 7-day SD newborn rats(weight>12g) divided into group①:normal control group and group②:sham-operated control group randomly by block. The remaining rats operated to build HIBD model, the success model divided into group③:model control group, group④:acupuncture group, group⑤:acupuncture + NGF group randomly by block.2. Intervened,weight,over hanging test,inclined plane test,Y maze tests the experimental rats according to plans.3. Detect expression of brain nerve cell BCL-2,BAX and Casp3 mRNA by using fluorescence quantitative PCR (SYBRGreen I);observe pathological changes in hippocampus by using HE staining;count brain normal nerve cells;detect nerve cell cell apoptosis by using TUNEL.Results:1. Brain tissue in general:Therea are 33 rats successful modeling which can not turn over,balance dysfunction or left-handed rotation in 36 rats survival after modeled. when we drawn after the experiment,we can see: group③appeared large brain area tissue necrosis and vacuoles; group①and group②nothing abnormal detected; group④and group⑤appeared slight brain tissue necrosis and vacuoles.2. Weight gain: Compared with group③, in group①,group②,group④and group⑤,the mean of weight and the tendency of weight gain is higher,the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.3. OHT: Compared with group③, in group①,group②,group④and group⑤,the mean of 16d after operation OHT results and the tendency of strength improve is higher,the difference had statistical significance.Compared group⑤with group④, the difference had statistical significance.4. IPT: Compared with group③, in group①,group②,group④and group⑤,the mean of 16d after operation IPT results is lower and the tendency of coordination ability enhance is higher , the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.5. Y maze test: Compared with group③, in group①,group②,group④and group⑤,the mean of 18d after operation Y maze test results is higher,the difference had statistical significance.Compared group⑤with group④, the difference had statistical significance too.6. Expression of BCL-2:Compared with group③, in group①,group②,group④and group⑤,the mean of BCL-2 A value is higher,the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.7. Expression of BAX: Compared with group③, in group①,group②and group⑤,the mean of BAX A value is lower,the difference had statistical significance.Compared group④with group③, p=0.087,the difference had no statistical significance.8. Expression of Casp3:Compared with group③, in group①,group②,group④and group⑤,the mean of Casp3 A value is lower,the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.9. Change of pathological feature: Compared with group③, in group①,group②,group④and group⑤,the quantity of degenerative nerve cells in hippocampus is lower and the pathological feature ameliorate significantly.10. Quantity of normal nerve cells:Compared with group③, in group①,group②,group④and group⑤,the mean of normal nerve cells quantity is higher,the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.11. Apoptosis index of cortical cells:Compared with group③, in group①,group②,group④and group⑤,the mean of cortical cells apoptosis index is lower,the difference had statistical significance.Compared group⑤with group④, the difference had no statistical significance.Conclusion:1. Effect of Acupuncture on weight gain,muscle growth,coordination of recovery,learning and memory (mental) recovery of HIBD CP model rats played a significant role.2. Acupuncture can inhibit mRNA expression of apoptosis gene BAX and Casp3 and promote mRNA expression of anti-apoptosis gene BCL-2 in HIBD CP model rats obviously.3.Acupuncture can promote the injured nerve cells recovery and inhibite it apoptosis in HIBD CP model significantly.4.Nerve growth factor can play a synergistic effect with acupuncture on the treatment of CP in HIBD CP model.
