Objective The study investigated the relationship between cognitive injure of electrical kindled rats and expression of ChAT and 5-HT in hippocampus.Methods We established electric chronic kindled rats epilepsy model. Rats were divided into KE(kindled group), SHAM(operative not stimulate), and Control(not treated) groups randomly. After rats were kindled, they were divided into 7d,15d,30d,60d subgroups. EEG was recorded after rats were electric kindled when the rats were awake and freedom. EACh group were performed Morris water maze (MWM) test, first place navigation test (PNT) and then spatial probe test (SPT), to test the learning and memory retention ability. Perfuse and fix the rat brains and Nissl Staining were made to see the injure of neurons in hippocampus. Immunostaining of ChAT and 5-HT in hippocampus sections to investigate the relationship between their expression and cognitive injure.Results We could establish kindled epilepsy model by electric stimulate basal lateral amygdala . In MWM trial, 7 days after models were established, there is learning and memory retention disturbance in KE group compared to SHAM and Control group. It took longer time in PNT and few frequency in SPT in KE group than that in SHAM and Control group at 7d and 15d after rats were kindled(P<0.05). 30d and 60d there was no significant difference in PNT and SPT. Nissl staining displayed neurons in CA1,CA3 and dentate gyrus were deformed , cellular swelling, derangement etc. Expression of ChAT and 5-HT in KE was higher than that in SHAM and Ctronl group at 7d and 15d after kindled (P<0.05). No significant difference was found at 30d and 60d after rats were kindled.Conclusion We can established epilepsy model by electric stimulating basal later amygdale. Cognitive function was injured at early time of epilepsy. With the time going, few seizure frequency, cognitive was recovered to a certain extent. Cognitive injure of epilepsy has some relationship with neurons injury in hippocampus. There is relationship between expression of ChAT and 5-HT and cognitive injury. ChAT and 5-HT maybe the pathology foundation of cognitive injure.
