Hawthorn leaves flavonoids as modle drug,Hawthorn leaves flavonoids self- Microemulsifying membrane controlled-release coated droppill was prepared in this experiment by self-emulsifying technology and the sustained released coating technology, The release rate in vitro and Pharmacokinetics study in vivo following a single oral administration to rats were investigated.1,The preparation of the Hawthorn leaves flavonoids Self- microemulsionUsed self- Microemulsifying technology to prepare the Hawthorn leaves flavonoids Self- microemulsion, which was in order to improve the dissolution of Hawthorn leaves flavonoids. The basic components of the formulation were determined by investigating the solubility of Hawthorn leaves flavonoids in different emulsifiers and auxiliary emulsifiers, and the interaction between oils, emulsifiers and auxiliary emulsifiers. The pseudorernary phase diagram was drawn to determine the final components of the formulation. And a series of properties of the Hawthorn leaves flavonoids Self- microemulsion were investigated.2. The preparation of Hawthorn leaves flavonoids self- Microemulsifying droppill:Hawthorn leaves flavonoids self- Microemulsifying droppill was prepared with the PEG-6000, in the basic of Hawthorn leaves flavonoids Self- microemulsion which had been prepared above.3. The preparation of Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill:Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill was prepared by the coating technology, in the basic of Hawthorn leaves flavonoids Self- microemulsion which had been prepared above. The components of the coating liquid were as follows: Ethylcellulose(EC) was the coating material, PEG-400 was the porosity-making agent, DEP was the plasticizer. The releasing of droppill was controlled by controlling amount of EC, PEG-400, DEP and the Film Thickness. After investigating the effect of single fator,the optima1 formulation was obtained by uniform design.4. the study on vitro release rate of Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill:To evaluate the vitro release rate of Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill,The visible absorption spectroscopy and HPLC analytic methods were established. The vitro releasing curve of Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill was most qualified the first-order equation, and the reproducibility of releasing appeared good.5. The study on Pharmacokineties of Hawthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppill in rats:The SD rats were as the experimental animals. The SD rats were received intragastric administration with the awthorn leaves flavonoids self- Microemulsifying Membrane Controlled-release coated droppills which were qualified in vitro release rate, and Serial blood samples were collected at designed time points. HPLC was used for analysis of blood samples after they were purified. Pharmacokinetics parameters were calculated using 3P97 pharmacokinetic program, and a series of Pharmacokinetical paramrters,such as, t(peak),c(max),AUC, and so on, were got.
