The Studies On The Mechanism Of The Treatment On Rats With Prostatic Hyperplasia And The Effects Of The Expression Of PCNA , Bcl-2 , Bax By ZeGuilongShuang

ObjectiveProstate index change in pathology and the expreesion of PCNA Bcl-2 and Bax were surveyed to observe mechanism of action that ZeGuiLongShuang (ZGLS)treat rat Benign Prostate Hyperplasia(BPH).MethodsSD rats were divided into six groups at randomly: the sham operation group, the model group, the proscar group, the ZGLS high doses group, the ZGLS middle doses group, the ZGLS low doses group. The preparation of the prostatic hyperplasia model is induced by castrated rats with male testosterone induction method. The prostate index in BPH rats is measured. To observe the pathological change of prostatic hyperplasia in rats by HE dyeing. To observe PCNA,Bcl-2,Bax protein expression of prostate tissue by immunohistochemistry method.Results1,The effect of humid weight and prostate index in each groupFinasteride group with the same infirmity ,The influence about prostate humid weight and prostate index in each group:ZGLS can be cut down obviously SD rats of prostate wet weight and prostate index (P<0.05).2,The influence about prostate pathology tissue in each groupThe model group: Prostate glandular organs are tight arrangement, cellula epithelialis are high pillar.Cellula epithelialis are more than one unilayer,most of cellula epithelialis form convex papilate. There is blood vessel dilatation.The proscar and ZGLS group: Convex papilate are less than model group, lumen of gland become big,cellula epithelialis are unilayer pillar,there is no excreta in glandular cavity.There is some blood vessel dilatation. Morphological changes of the middle doses group, a high doses of proliferation in the heavier but lighter than the model group.3,The influence about the expression of PCNA Bcl-2 and BaxThe expression of PCNA is lower in each doses group of ZGLS and proscar group, compared with model group, there is significant difference(P<0.01). The expression of Bcl-2 is lower in each doses group of ZGLS and proscar group, compared with model group, there is significant difference(P<0.01). The expression of Bax is higher in each doses group of ZGLS and proscar group, compared with model group, there is significant difference(P<0.01).ConclusionsSubcutaneous injection of testosterone propionate rat BPH model manufacturing, each ZGLS group can reduce wet weight of rat prostate BPH, prostate index decreased to treatment of benign prostatic hyperplasia, especially high doses of ZGLS group cool effect and no significant difference between finasteride. The possible mechanism of treatment of BPH with ZGLS is by reducing the PCNA, Bcl-2 expression, increasing expression of Bax, to decrease the proliferation activity of glandular cells while promoting apoptosis.
