ObjectiveTo detect the site of ACE in kidney of normal rats and models rats. Determination of type 2 diabetic nephropathy (DN) occurred in the process of the development of blood,Urine angiotensin-converting enzyme (ACE) and discuss its clinical significance.MethodsWistar rats with diabetes induced by streptozotocin(STZ) were sacrificed after injection of STZ. using the rapid blood glucose meter measuring blood sugar after 72 hours,≥16.65 mmol/L of the blood sugarinl experimental rats, The control group inject equal doses of folic acid buffer fluid. Five rats inject STZ broken neck after first 60 days,control group three rats. Cut Kidney parts ganiza- tion to make specimen. Using RIA chemical theory, Perfusion fixed, Frozen section, fixation,imaging, observation ACE in normal and model mice kidney. Using rate method detection 65 cases of type 2 diabetes patients and 50 cases of normal healthy human blood,urine ACE activity and urine ACE/(Cr) ratio, simultaneous determination of microalbuminuria (mALB), urinary N-acetyl -beta D amino glycosidase enzymes were identified (NAG) level and they and the ratio of serum creatinine (Cr), comprehensive analysis of test results.ResultsType 2 diabetic patients with urinary ACE, mALB, NAG was significantly higher than the control group, the difference was significant (P<0.05), 2 type diabetic patients with urinary ACE,the difference in each group was significant (P<0.01). urinary ACE was no correlated with serum ACE ,on the other hand, urinary ACE was correlated with mALB,NAG.Urine ACE and urine ACE/urine Cr higher than mAlB,urinary NAG group in type 2 diabetic patients. The sensitivity of type 2 diabetic nephropathy 83%, 90% , specificity 73%, 86%.that urinary ACE activity progressively increases during the development of early diabetic Nephropathy. ACE exists in renal cortex ,the renal cortex of Normal rats with a small amount of the ACE, the renal cortical tubules of model group rats are seen large ACE.ConclusionsRIA has appeared in organization with precise position, image clear, provided an objective basis for the clinical significance. Detection and comparative analysis of type 2 diabetic patients with urinary ACE activity, as well as mALB, NAG,serum ACE for early renal damage in type 2 diabetes, Illustratesd ACE activity in urine has some clinical value.
