ObjectiveThis experiment prepared for rabbit VX2 implantation tumor model of breast, using the f-MSCT(functional multi-slice computed tomograph) to evaluate anti-angiogenic effect that treated by Endostar with Radiation Therapy, comparing pathology to investigate the mechanism of this combination therapy, and it can supply experimental basis for clinical applicati treatment in breast cancer, and further assessment to find.Programs of Endostar combined with radiation therapy in anti-angiogenic effect of treatment methods.Methods64 Japan white rabbits(in 3-4 months and 2.0-2.5kg) successfully implanted VX2 tumor in the unilateral brest were divided into groups at random, treatment divided into 4 groups(N=16),which divided into Saline group, anti-angiogenic group, radiotherapy,radiation combined with antiangiogenic therapy group, after 2 weeks of tumor types, treated tumor-bearing rabbits underwent GE Light speed 16 spiral CT perfusion imaging, Observing perfusion maps, using the deconvolution model of perfusion 3 software to calculate the value of the three perfusion parameters ;the blood flow (BF), blood volume (BV) and permeability surface (PS); after that the group of anti-angiogenic monotherapy was given Endostar;radiation treatment group received radiotherapy alone; combined treatment group were treated by Endostar with Radiation Therapy two programs. Each group underwent CT perfusion scan after treatment , observing Perfusion maps and analysing datas,while removing fresh tumor tissue, detected by immunohistochemical expression of VEGF. Application package by SPSS13.0 t test, variance analysis and correlation analysis methods for statistical analysis, P <0.05 level was significantly.ResultsThe mean values of CT perfusion parameters BF,BV and PS in tumor tissue were higher than those in the muscle tissue at the same slice before therapy, There was no significant difference between each group (P> 0.05); after treatment Endostar group, radiation therapy group and combined treatment group of CT perfusion parameters BF, BV, PS were significantly lower than before treatment ,meaning differences were statistically significant (P <0.01), Saline group compared with a rise above parameters (P <0.01); Each group BF, BV, and PS mean pairwise comparisons were significantly different (P <0.01), showing the lowest group of Combined treatment ; radiation therapy was centered; Endostar group higher, but lower than the control group, saline was the highest, the role of combination therapy was the most, Endostar and radiation therapy in combination have a synergistic effect . CT perfusion parameters of BF, BV, and PS had a positive correlation with VEGF expression in experiments each group VX2 rabbit mammary tumors.Conclusions1,Endostar monotherapy planted VX2 tumor in rabbit mammary gland may play a role in anti-angiogenesis that can reduce VEGF expresion of tumor significantly; radiation therapy in rabbit VX2 breast tumor can make anti-angiogenesis effect also; Endostar in combination with radiation therapy had a synergistic effect on treatment of rabbit VX2 implanted tumor in the breast .2,f-MSCT can indirectly reflect the growing rabbit VX2 tumor Endostar breast,both chemotherapy and radiotherapy of tumor angiogenesis and immunohistochemical of VEGF expression changes after treatment,f -MSCT can be used as in vivo, noninvasive evaluation of Endostar, radiation therapy and two programs combined treatment of rabbit VX2 breast tumor anti-angiogenesis effect of planting method.
