Objective:To study the expression levels of Sambucus Nigra Agglutinin(SNA)N Solarium Tuberosum(Potato)Lectin(STL),Soybean Agglutinin(SBA) and their clinicopathological significance in the margin and malignant lesions of stomach.Methods:Immunohistochemistry for determining SNA,STL,SBA expression level were used in conventional paraffin-embeded sections from specimens of gastric caner and correlative margin lesions(n=60).Results:1.Both gastric carcinoma regions and margin gastric mucosa regions showed SNA,STL,SBA-positive staining.Positive staining was located on the cell surface and cytoplasm. In areas of gastric margin lesion,positive staining of STL was mainly located on the gland cell(51.9%)(P<0.01);positive staining of SBA was mainly located on the epithelium cell(55.1%) (P<0.01).2.The positive rate of SNA was significantly higher in gastric cancer tissues than those in margin tissues (P<0.01).The positive rate of SNA was significant higher in poorly-differentiated,metastasis of regional lymph node and worse TNM staging than those in well-differentiated,no-metastasis of regional lymph node and better TNM staging in gastric cancer (P<0.05).In the regression analysis,the positive of SNA was significant higher in worse TNM staging than those in better TNM staging in gastric cancer (P<0.05).3.The positive rate of STL was significantly lower in gastric cancer tissues than those in margin tissues (/)<0.01).The positive rate of STL was significant higher in poorly-differentiated and worse TNM staging than those in well-differentiated and better TNM staging (P<0.05).In the regression analysis,the positive of STL was significant higher in poorly-differentiated and worse TNM staging than those in well-differentiated and better TNM staging in gastric cancer (P<0.05).4.The positive rate of SBA was significantly lower in gastric cancer tissues than those in margin tissues (P<0.01).The positive rate of STL was significant higher in poorly-differentiated,deep infitrating depth and worse TNM staging than those in well-differentiated,superficial infitrating depth and better TNM staging (P<0.05).In the regression analysis,the positive of SBA was significant higher in deep infitrating depth than superficial infitrating depth in gastric cancer (P<0.05).5.In the gastric cancer,the spearman rank correlation test showed that STL expression was closely associated with SBA (r=0.266,P=0.020) and SBA expression was closely associated with SNA (r=-0.266,P=0.020).The significant difference was found in different infitrating depth differentiation grade,metastasis of regional lymph node and TNM staging when analysing distribution of STL and SBA (P<0.05).Conclusion: 1.Both gastric carcinoma regions and margin gastric mucosa regions showed SNA,STL,SBA-positive staining.Positive staining was located on the cell surface and cytoplasm.In areas of gastric margin lesion,positive staining of STL was mainly located on the gland cell(51.9%)(P< 0.01);positive staining of SBA was mainly located on the epithelium cell(55.1%) (P<0.01).2.The expression of SNA was significantly correlated with differentiation grade,lymphnode metastasis and TNM staging (P<0.05).The expressive level of SNA may be important molecular markers of lectins for reflecting the carcinogenesis, progression and biological behaviors in gastric cancer.3.The expression of STL was significantly correlated with differentiation grade and TNM staging (P<0.05).The expression of SBA was significantly correlated with differentiation grade infitrating depth and TNM staging (P<0.05).High levels of STL and SBA seem to be closely associated with favorable behaviors of human gastric adenocarcinoma.STL and SBA might be a useful pathological index for diagnosis and prognosis of human gastric adenocarcinoma.4.In the gastric cancer,the spearman rank correlation test showed that STL expression was closely associated with SBA (P=0.040, r=0.266) and SBA expression was closely associated with SNA (P=0.040, r=-0.266).
