| Objective:To investigate the function of heaptocyte nuclear factor 6 (HNF-6) during the early stage of liver regeneration after the partial hepatectomy of rat.Methods Using Higgins'method,and 30 SD rats were taken to construct the liver partial hepatectomy model of rat. Then the morphological changes of normal liver and the residual liver was observed.Immunohistochemistry assays and RT-PCR were used to determine the expressing changes of HNF-6 during the early stage of liver regeneration.Results:â‘ he morphological changes of rat's liver during the early stage of liver regeneration:The main morphological changes of liver during the early stage of liver regeneration is hyperemia and edema, the broadening of hepatic blood sinus and the structural disorder of hepatic cords and hepatic lobule,and inflammatory reaction were also seen among liver cells.Along with the progress of the regeneration process, hepatic blood sinus further broadened. In addition, the number of hepatic cells and bile duct epithelial cells increased, dual-core phenomenon and visible diffuse cavitation were seen among liver cells.â‘¡The expressing of HNF-6 during the early stage of liver regeneration:Immunohistochemical assays demonstrated that the espressing of HNF-6 in normal hepatic was weak,but its espressing in normal bile duct epithelial is obvious. Its expressing natably increased in all the experimental groups. The espressing places of HNF-6 were located in the nucleus and cytoplasm, and it's expressing reached it's peak at 6h after PH, Then, it's expressing gradually dropped, but it was still higher than normal group.â‘¢The RT-PCR result of HNF-6: RT-PCR assays demonstrated that HNF-6 expressed during the all early stage of liver regeneration and in normal hepatic cell.Conclusions HNF-6 may regulate the starting of liver regeneration, and this founction of it may be in connected with its interaction with cyclin factor. |
