Expression Of HDAC6 In Esophageal Carcinoma And Its Clinical Significance

Objective:To study the expression of HDAC6 levels in esophageal squmous cell carcinoma (ESCC), the correlation between expression of HDAC6 in ESCC, and relationship with clinicopathologic characters, such as depth of invasion, lymph node metastasis, TNM stage and histologic grade, and to investigate its role in tumorigenesis as well as progression in ESCC and their clinical significant.Method:The expression of HDAC6 was detected in 50 cases of esophageal squamous cell carcinoma tissues and the 30 cases of para-cancer normal esophageal tissues by immunohistochemistry and their relation to clinical pathologic characteristics of ESCC was analyzed.Result:1. In ESCC cells, HDAC6 revealed a predominantly cytoplastic localization and by immunohistochemical analysis, demonstating that the expression of HDAC6 in cancer is significantly higher than low expression that in the para-cancer normal esophageal tissues.The expression rate of HDAC6 in ESCC (82.0%,41/50) was significantly higher than that in the para-cancer normal esophageal tissues 56.7% (17/30) (x2=6.036, P=0.014<0.05)2.By immunohistochemical analysis, HDAC6 was expressed significantly higher in lymph node metastasis tumors than that in lymph no node metastasis (x2=13.375, P=0.008<0.05),in late-stage(III,IV) metaphase of TNM stage(TNM classification of esophageal carcinoma-7th edition) tumors is more than that in (Ⅰ,Ⅱ)early -stage (x2=4.818, P= 0.028<0.05).However, divide depth of invasion,histologic grade,Age into two grade (T1/T2 and T3/T4, high-middle and low differentiation,≥60 and<60 years), HDAC6 had no correlation to age, depth of invasion, carcinoma differentiation.Conclusion:HDAC6 revealed a predominantly cytoplastic localization In ESCC cells and by immunohistochemical analysis,demonstrating that the expression of HDAC6 in cancer is significantly higher than low expression that in the para-cancer normal esophageal tissues, HDAC6 was expressed significantly higher in lymph node metastasis tumors than that in lymph no node metastasis.The expression of HDAC6 correlated with lymph node metastasis and TNM stage.Indicating HDAC6 protein is related to ESCC development and tumor headway,metastasis. HDAC6 maybe act as one of biological indicators of tumor headway and metastatic potential and medicine target spot in diagnosis and therapy of ESCC.
