| To illustrate the effect of Puerarin on progesterone (P) and estradiol's (E2) levels in mice serum and on progesterone receptors (PR) and estrogen receptor's (E2R) gene expression's dynamic changes in breast tissue, the test divided 240 mice of 21 days old into four groups, as the control group, low dose of Puerarin, middle dose of Puerarin and high dose of Puerarin, each groups has 60 mice, Administered once every other day. Control group was given 0.2ml of distilled water, the low dose group was given 0.2ml of 0.5mg/ml Puerarin solution, the dose group was given 0.2ml of 1.0mg/ml Puerarin solution, high dose group was given 0.2ml of 1.5mg/ml Puerarin solution, respectively in the 40,46,52,58,64,70 day killed the mice in each group by cervical and then collecte the blood and take the breast, using radioimmunoassay and real-time quantitative RT-PCR detected the progesterone (P), estradiol's(E2) levels in serum and progesterone receptors (PR), estrogen receptor's (E2R) expression in breast tissue.The results showed:1,there's no effect of low, medium and high dose of puerarin group on progesterone of different days hormone in mice serum.2,low dose of puerarin can significantly increased the estradiol levels on 70 day in mice;the medium dose of puerarin can significantly increased the estradiol levels start in 52 days;64 days of age decreased,70 days significantly improved again;high dose of puerarin had no significant effect on estradiol in other day's mice,but in 70 days it feedback inhibition the estradiol's secretion trends.3,low, medium and high dose of puerarin group increased from 52 days of age began to increase with age increasing trend on progesterone receptor's expression.4,low doses of puerarin begin to significant improve the estrogen receptor expression in mice on 58 days;middle and high dose of puerarin groupincreased from 52 days of age began to increase with age increasing trend on estrogen receptor's expression.
