ObjectivesTo investigate the expression of TNF-αand IL-2 in acute rejection response following liver transplantation, and evaluate their roles to be used as markers of acute rejection.MethodsBy establishing orthotopic rat liver transplantation models, Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the gene expression of TNF-αand IL-2 in the graft for diagnosis of acute rejection after liver transplantation in rats. The diagnosis of acute rejection was based on histopathology. The relationship between the TNF-α/IL-2 expression and acute rejection was analyzed.ResultsThe mean levels of TNF-αand IL- 2 in liver tissues on days 3,5and 7 were significantly higher in allogene group than those of isogene group.ConclusionsTNF-αand IL-2 expression seemed to be related to the development of acute rejection following liver transplantation,and might be served as sensitive and reliable markers in diagnosing acute rejection in early stage.
