Objective: To observe the effect on the nerve growth factor (Nerve growth factor, NGF) ,nerve growth associated protein -43 (Nerve growth associated protein-43) mRNA and protein expression of the F1 generation rat hippocampus morphology after exposure to sodium arsenite during pregnancy and lactation, and explore the mechanism of central nervous system damage of F1 generation rat after exposure on maternal arsenic.Methods: 12 of pregnancy female rats on first 0 days, randomly divided into 4 groups, each group have 3 rats, toxin exposure groups are 10mg/L, 50mg/L, 100mg/LNaASO2 dose and control group, from the sixth day began to drink gestation, continuous most poisonous arsenic until 21 days after offspring was born.Afterwards, sacrificed the female mice for experiments. Observe the change of form and numbers in CA1,CA3,DG area neurons in hippocampus by HE and Nissl stain, Immunohistochemical was used to observe NGF, GAP-43 protein expression in CA1, CA3, DG area of each rat hippocampus; We extracted the NGF,GAP-43mRNA on the rat hippocampus tissue for RT-PCR and use Western blot to measure the NGF,GAP-43 protein expression.Results: HE stain showed the control group the hippocampus neurons rich integrity in CA1,CA3 area, edge is clear, the nuclei are round, oval, in nuclear chromatin are uniformly distributed, nucleoli is clear. The dye arsenic group visible cultured hippocampus neurons edge owe clear, cell bodies shrinks, nuclear crimp and scattered phenmomenon, such as high dose group of apoptosis be more serious.Nissl stain: control group hippocampus Nebuchadnezzar neuronal cell cytoplasm with rich bartonella, with the poisonous doses increasing, bartonella decreasing. The most obvious is high-dosage group, successively are medium-dose group and low-dose group, and most obvious is CA1 area damage.Immunohistochemical tests showed the average light density of GAP– 43, NGF in the, high, medium and low dose group is lower than the control group, (P < 0.001, P < 0.001). but the differences in hippocampal DG district have no statistically significant compare with control group. (P > 0.05).RT - PCR results show that the NGF, GAP - 43 mRNA expression reduced, and has significant difference with control group. (P < 0.001, P < 0.001).Western blot results show that the NGF, GAP - 43 mRNA expression reduced has significant difference with control group. (P < 0.001, P < 0.001). Conclusion: Rats exposure to arsenic during Pregnancy and lactation can damage hippocampus neuronal cell, restrain the NGF, GAP - 43mRNA and protein expression of F1 generation rat, suggest that rat exposure to arsenic during pregnancy and lactation can damage hippocampus issue form, reduce the expression level of NGF,GAP-43 in the hippocampus issue, such mechanisms could create damage on the central nervous system of the F1 generation rat.
