PARTâ… Animal model establish and protective effects of curcumin on neonatal rats with Bronchopulmonary DysplasiaObjective: To establish animal model of hyperoxia-induced bronchopulmonary dysplasia(BPD)in neonatal rats,determine the effect of prolonged hyperoxia on the development of rat lung,and evaluate protective effects of curcumin on neonatal rats with BPD.Methods: Eighty SD rats born within 6h were randomly divided into 4 groups: room air group(Groupâ… ), hyperoxia group(Groupâ…¡), hyperoxia + vehicle group(Groupâ…¢), hyperoxia + curcumin group(Groupâ…£). The general situations and weights of the rats were observed,Pathomorphology of the lungs was observed with hematoxylin-eosine staining on day 4 and 14.Results: Compared with the normoxic group, rats exposed to hyperoxia showed a histological pattern of alveolar simplification characterized by the presence of larger and fewer distal air spaces, Curcumin treatment ameliorated the histopathological signs. In addition, the radial alveolar counting (RAC) was significantly lower, which was significantly attenuated with curcumin treatment for 14 days.Conclusions: Prolonged hyperoxia exposure appears to lead to arrest of lung development. In addition, the radial alveolar counting (RAC) was significantly lower, which was significantly attenuated with curcumin treatment for 14 days. Hyperoxia exposure also resulted in significant cell apoptosis, which was reduced after treatment with curcumin. Both Immunohistochemistry and Western blot demonstrated increased expression of activated caspase-3 in the lung tissue after hyperoxia exposure, which was reduced by curcumin treatment for 4 days.Conclusions: Cell apoptosis in involved in the formation of bronchopulmonary dysplasia. Curcumin has protective effects against BPD in neonatal rats, possibly through inhibiting the expression of active Caspase-3 and reducing cell apoptosis in lung tissue.
