Objective: This study was to investigate the effect of ischemia pretreatment on cold ischemia kidneys for transplantation. The thesis contained: the effect of ischemia pretreatment on pH value and lactate dehydrogenase activity in preservation solution, and the extent of oxidative stress in renal tissues; and the possible role of heme oxygenase-1 in ischemia preconditioning.Method: (1) Measuring pH value and lactate dehydrogenase activity in preservation solution were adopted to investigate the effect of ischemia pretreatment on cell injury in cold ischemia kidneys. (2) The extent of oxidative stress was assessed by detection of renal reduced glutathione and malondialdehyde contents. (3)Western blotting method was used to measure the expression of heme oxygenase-1 in ischemia pretreated kidneys with or without cold ischemia preservation.Result: (1)pH values in preservation solution at 24h, 48h, 72h after ischemia pretreatment were higher than that of the group without ischemia pretreatment. The pH values in the group with heme oxygenase-1 inhibitor administration obtained no statistic significance in comparison with the control group. (2) The lactate dehydrogenase activities in preservation solution at 48h, 72h after ischemia pretreatment were lower than that of the group without ischemia pretreatment. This effect was not obtained after the administration of heme oxygenase-1 inhibitor. (3) The content of renal reduced glutathione in ischemia pretreatment group at 24h and 48h were higher than that of the group without ischemia pretreatment, while the content of malondialdehyde were lower than the group without ischemia pretreatment at the same time point. (4)Ischemia pretreatment induced heme oxygenase-1 expression in rat kidney, and lasted to 96h after treatment, while the cold preservation treatment did not change this protein's expression.Conclusion: ischemia pretreatment effectively made improvement on cell injury and oxidative stress in cold ischemia rat kidneys, and the action of ischemia pretreatment may be contributed to the induction of heme oxygenase-1 via its anti-oxidative property, while other actions such as anti-apoptosis role can not be ignored and remain to be elucidated in the further investigation.
