Objective:We designed a series of experiments to investigate the role of epidermal growth factor (EGF)/ mitogen-activated protein kinases (MAPK)signaling pathways in the regulation of matrix metalloproteinase 9(MMP-9)in JEG-3 trophoblast cells.Methods:The JEG-3 trophoblast cell line was used in this study. The expression of MMP-9 in the trophoblast cell line stimulated by EGF was tested by Western blotting and Reverse transcription polymerase chain reaction (RT-PCR), respectively. The activity of MAPK (P38,ERK) signaling pathway in EGF-stimulated cells was measured by Western blotting. The effects of the MAPK (P38,ERK) pathway specific inhibitors(SB203580,U0126)on EGF-induced MMP-9 expression in JEG-3 cells were examined by Western blotting.Results:(1)EGF induced expression of MMP-9 in a time and dose-dependant manner in JEG-3 cells(.2)P38 and ERK were activated in JEG-3 cells by EGF(.3)SB203580 and U0126,specific blocker for P38 and ERK , significantly inhibited the production of MMP-9 and NF-КB by EGF.Conclusion:EGF induced expression of MMP-9 in a time and dose-dependant manner in JEG-3 cells;EGF induced enhanced MMP-9 expression mediated by P38 and ERK signaling pathway in JEG-3 trophoblast cells.
