| Objective: Coronary heart disease is prevalent in modern society. The pathogenesis of artherosclerosis is a complicated procedure with multiple factor participating. While there are many theories about artherosclerosis, the details of its mechanism are still unknown. Recently, the epigenetic abnormal-regulator of gene expression was found to play an important role in the atherosclerosis pathogenesis. The present study was design to discuss the relationship among methylation of ER-αgene CpG island , mRNA expression and coronary atherosclerosis by using Methylation-specific polymerase chain reaction and Real-time RT-PCR.Methods: The blood samples from 40 patients with coronary atherosclerosis and 20 healthy control patients were collected in Anhui Provincial Hospital. Genomic DNA was extracted from blood by using kit, and then treated with bisulfite. The methylation-specific PCR (MSP) was adopted to detect the promoter CpG islands hypermethylation of ER-αgene in the above-mentioned two kinds of specimens. The mRNA expression was investigated by The Real-time RT-PCR.Results: (1) The positive rates of hypermethylation on promoter region of ER-αgene was 77.5% in CHD group and 55% in control group, P<0.05. (2) The mRNA expression in negative samples were much higher than that in positive samples, while the multiple of the differences between CHD group and control group was 2.57, and there was a significant descend in CHD group. (3) There were much more samples got hypermethylation on promoter region of ER-αgene in old patients compared with the young patients.Conclusion: The aberrant methylation of ER-αis one of the important mechanism leading to the low expression of mRNA. And this change may contribute to the development of the coronary atherosclerosis. |
