Objective: Endometriosis(EMs, referred to as endometriosis ) is a kind of disease that refers to growth function of endometrial glands and stroma cells are present in outside of normal uterus gut lining, is a common illness with the women in reproductive year and the trend of morbidity has increased year by year. Dysmenorrhea, persistent next bellyacke, menstruation and infertility are the common clinical manifestations of Ems patients. But so far, the exact mechanism has not fully understood. Although Ems is a benign lesion, its extensive cultivation, easy relapse characteristics are similar to malignant tumors. Recent research shows that: the lack of oxygen in malignant tumors are playing an important role in the process. Recent research shows that EPO has the role in promoting new angiogenesis; HIF-1 is the core regulatory factors of anoxic condition angiogenesis, and the relationship with angiogenesis has been confirmed by a large number of studies, it can be influenced by other expression of cytokines participate in the whole process of angiogenesis. Clinical treatment with Ems western medicine mainly adopts different surgical treatment, drug therapy, expect therapy. The chinese medicine widely adopts more oral medicine herbs in enema synthetic therapy. This topic plans through animal experiments to study the relationship lack of oxygen and the occurr of Ems, in order to explore the part pathogenesis of Ems; And the method of using Soothing liver regulating qi warming yang and removing blood stasis to treat erperimentof rats, discusses the effect and function mechanism, to provide a new thinking for treatment.Method: 60 depuratory and healthy wister females rats with normal strous cycle, 200±20g in body weight, 10 is divided into normal group, others are used wangyanping method to duplicate the model of endometriosis. After 4 weeks, chose the 40 rats that duplicate model triumphantly and divided them into 4 group: high-dose group, low-dose group, model group, Danazol group. There were 10 rats in each group. Those rats of low dose and high dose groups were administrated in dose of 15.41g/kg,61.76g/kg separately, once per day for 4 weeks. The rats of Danazol group were in dose of 100mg once daily. Model group and normal group were given the same voluminal distilled water. The last time after treatment, transvaginal smear, checked estrogen cycle, chose estrogen period of rats, fasting eight hours after last administration, sacrificed the rats by decapitation during anestrum, took ectopic endometrium caesarean organization of each guoup and the normal group took endometrial organization, blotting blood after, Fixed endometrium in 4% Polyoxymethylene, conventional paraffin for embedding, slicein 5μm. Carried out immunohistochemical staining, determination the expression of EPO, HIF-1 in endometrial tissue.Result:1 EPO positive expression mainly in graft ectopic endometrium glands cell cytoplasm, nuclear and cytoplasmic with small. High-dose group, low-dose groups were lower than model group, the two groups had significant differences with model group(P<0.05), high-dose group and low-dose had no significant difference(P﹥0.05); model group was higher than normal group, there were significant differences between the two groups(P<0.05); high-dose group, low-dose and normal group had no difference(P>0.05); high-dose group lower than western medicine group, there were significant differences between the two groups(P<0.05), low-dose and western medicine group had no difference(P﹥0.05).2 HIF-1 positive expression in transplant ectopic endometrium glandular epithelium cells in the cytoplasm, individual ectopic endometrium stroma with small expressionm. High-dose group, low-dose groups were lower than model group, the two groups had significant differences with model group(P<0.05), high-dose group and low-dose had no significant difference(P﹥0.05), model group was higher than normal group, there were significant differences between the two groups(P<0.05); high-dose group, low-dose and normal group had no difference(P>0.05); high-dose group lower than western medicine group, there were significant differences between the two groups(P<0.05), low-dose and western medicine group had no difference(P﹥0.05).Conclusions:1 The ectopic endometrium organization fragments of EMs occurred planting disseminated is in ischemia, anoxic condition, this state by inducing ectiectpic endometrium cells EPO, HIF-1 and down-regulate then promote the formation of new blood vessels local groups.2 High expression of EPO in experimental rats ectopic tissues help generate new vessels, promote the occurrence of EMs.3 High expression of HIF-1 in experimental rat ectopic tissues indicates that ectopic tissue in the hypoxia phenomenon, hypoxia may be a main cause for EMs.4 Animals were treated by Shuganliqiwenyanghuayu chinese medicine, reduced the expression level of EPO, HIF-1, thus hint Shuganliqiwenyanghuayu method can inhibit EPO, HIF-1 expression, it plays a role in treatment of endometriosis.
